摘要:
目的:评价川芎平喘合剂对慢性阻塞性肺疾病(COPD)相关肺动脉高压大鼠肺动脉压的作用及对一氧化氮(NO)、内皮素1(ET-1)的影响。方法:将SD大鼠60只随机分为对照组、模型组、川芎平喘合剂组、法舒地尔组。采用烟熏联合脂多糖并给予低氧环境方法复制COPD相关肺动脉高压模型。采用多通道生物信号采集系统测定大鼠平均肺动脉压力,采用硝酸还原酶法测定NO,酶联免疫吸附法测定ET-1。结果:模型组平均肺动脉压(mPAP)高于对照组(P<0.01),法舒地尔、川芎平喘合剂组mPAP均较模型组下降(P<0.05);模型组、川芎平喘合剂组右心肥大指数(RVHI)高于对照组(P<0.05),川芎平喘合剂组、法舒地尔组RVHI与模型组比较差异无统计学意义(P>0.05);模型组较对照组NO水平降低,ET-1水平升高(P<0.01);川芎平喘合剂组、法舒地尔组较模型组NO水平升高,ET-1水平降低(P<0.05)。结论:川芎平喘合剂对COPD相关PAH具有治疗作用,作用机制与调节肺组织ET-1、NO表达有关。
Abstract:
Objective: To evaluate the effects of ChuanXiong asthma-relieving mixture on pulmonary arterial pressure of rat model with COPD associated pulmonary artery hypertension(PAH) and its influence on NO and ET-1. Methods: Sixty rats were separated into the control group, the model group, ChuanXiong asthma-relieving mixture group and fasudil group. COPD associated PAH rat models were depulicated by sootiness and lipopolysaccharide under low oxygen environment. Mean pulmonary artery pressure(mPAP) of the rats were detected by multichannel biological signal acquisition system, NO by nitrate reductase method and ET-1 by enzyme linked immunosorbent assay (ELISA). Results: The model group was higher than the control group in mPAP(P<0.01), mPAP lowered in fasudil group and ChuanXiong asthma-relieving mixture group compared with the model group (P<0.05); right ventricular heart index (RVHI) of the model group and ChuanXiong asthma-relieving mixture group was higher than that of the control group(P<0.05), the difference had no statistical meaning in the comparisons of RVHI when ChuanXiong asthma-relieving mixture group and fasudil group were compared with the model group; when compared with the control group, the level of NO lowered and the level of ET-1 raised in the model group(P<0.01); NO level rose while the level of ET-1 lowered in ChuanXiong asthma-relieving mixture group and fasudil group compared with the model group(P<0.05). Conclusion: ChuanXiong asthma-relieving mixture shows therapeutic effects for COPD associated PAH, and its mechanism might be related to regulating the expressions of ET-1 and NO in pulmonary tissue.
