摘要:
目的:研究大蒜素对慢性肾衰(CRF)大鼠肾脏组织的保护作用并探讨其机制。方法:将100只实验大鼠随机分为正常对照组、模型对照组,大蒜素低、中、高剂量组,每组20只。采用腺嘌呤溶液[(250 mg/(kg·d)]灌胃21天的方法建立CRF大鼠模型,大蒜素各组腹腔注射大蒜素[5、10、20 mg/(kg·d)]进行治疗,正常对照组及模型对照组给予正常饮食,疗程28天。观察各组大鼠一般生存状态,测定体质量、24小时尿量、24小时尿蛋白量,计算肾脏指数,测定血清尿素氮(BUN)、血肌酐(SCr)、尿酸(UA)含量;观察肾脏组织病理变化及细胞凋亡指数(AI),检测抗氧化酶[超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)]活性和丙二醛(MDA)含量。结果:与模型对照组比较,经大蒜素治疗28天能够有效改善CRF大鼠一般生存状态;大蒜素20 mg/(kg·d)组CRF大鼠体质量显著升高(P<0.05),大蒜素10、20 mg/(kg·d)组24小时尿量显著减少、24小时尿蛋白量显著降低(P<0.05),肾脏指数显著降低(P<0.05),血清BUN、SCr、UA含量显著降低(P<0.05);经大蒜素治疗能够明显改善CRF大鼠肾脏组织病变并抑制细胞凋亡,大蒜素10、20 mg/(kg·d)组AI显著降低(P<0.05),肾脏组织SOD活性显著升高,MDA含量显著降低(P<0.05),20 mg/(kg·d)组CAT、GSH-Px活性显著升高(P<0.05)。结论:大蒜素对CRF大鼠肾脏组织具有一定的保护作用,可能与其能够降低氧化应激损伤、抑制细胞凋亡有关。
Abstract:
Objective:To study the protective effects of allimin on renal tissue of the rats with chronic renal failure (CRF), and initially survey the mechanism. Methods: All 100 rats were randomized into normal control group,model control group,low,moderate and high dosages groups of allimin,20 rats each group,Intragastric administration of adenine solution [250 mg/(kg·d)] for 21 days was used to build CRF rat models, they were treated by peritoneal injection of allimin [5, 10 and 20 mg/(kg·d)], normal control group and model control group accepted normal diet,the course was 28 days. The contents of MDA, GSH-Px, CAT and SOD were detected, apoptosis index(AI) and pathological changes of renal tissue were observed; the contents of UA, SCr and BUN in the serum were measured, renal index was calculated, 24 hours urinary protein excretion(24 hUPE), 24 hours urinary volume and body weight were detected, general living state of the rats in dfferent groups were observed. Results: Compared with model control group, treating with allimin for 28 days could improve general living state of CRF rats effectively; body weight of CRF rats in allimin group [20 mg/(kg·d)] rose significantly (P<0.05), 24 h urinary volume of the rats in the allimin groups of 10 and 20 mg/(kg·d) reduced notably, and 24hUPE lowered remarkably (P<0.05), renal index reduced notably (P<0.05), the contents of BUN, SCr and UA decreased significantly (P<0.05); allimin could obviously improve the pathological changes of renal tissue of CRF rats and inhibit cellular apoptosis, AI lowered notably in the allimin groups of 10 and 20 mg/(kg·d) (P<0.05); the activity of SOD in renal tissue rose notably, and the contents of MDA decreased apparently(P<0.05), the activity of CAT, GSH-Px of the group of 20 mg/(kg·d) rose signficantly(P<0.05). Conclusion: Allimin possess certain protective effects to renal tissue of CRF rats, which might be related to decreasing oxidative stress injury and inhibiting cellular apoptosis.
