摘要:
目的：探讨中药复方抗瘤丸治疗神经胶质瘤的诱导凋亡作用机制。方法：制备裸鼠神经胶质瘤模型分为模型组、抗瘤丸高、中、低剂量组和阳性药(替莫唑胺)组，TUNEL染色法检测肿瘤细胞凋亡率，Western blot方法检测细胞凋亡相关蛋白(Bax、Bcl-2)的表达变化。结果：与模型组比较，给药20天后，抗瘤丸低剂量组和阳性药组肿瘤细胞凋亡率明显增加；与模型组比较，所有给药组均能明显下调Bcl-2的表达(P＜0.05)，抗瘤丸低剂量组，阳性药组能显著上调Bax的表达(P＜0.05)，各给药组Bcl-2/Bax的比值减小(P＜0.05)。结论：抗瘤丸可降低肿瘤凋亡蛋白比值，促进肿瘤细胞的凋亡。
Abstract:
Objective：To explore apoptosis-inducing mechanism of herbal compounds KangLiu pills in treating glioma. Methods: Nude mice models with glioma were prepared, and divided into the model group, high, moderate and low dose groups of KangLiu pills and positive medicine group (temozolomide capsules) in the light of the size of tumor, TUNEL staining method was used to measure tumor cells apoptosis rate, and Western blot method was adopted to detect the expressions of cells apoptosis -related protein (Bax and Bcl-2). Results: Compared with the model group, after medicating for 20 days, tumor cells apoptosis rates in low dose group of KangLiu pills and positive medicine group raised apparently; compared with the model group, all the medication groups could obviously downregulate the expression of Bcl-2 (P＜0.05), low dose group of KangLiu pills and positive medicine group could upregulate the expressions of Bax (P＜0.05), the ratio of Bcl-2/Bax reduced in all the medication groups (P＜0.05). Conclusion: KangLiu pills could reduce the ratio of tumor apotosis protein and promote the apoptosis of tumor cells.
