摘要:
目的:观察酸枣仁汤对慢性睡眠剥夺大鼠海马5-HT1AR和5-HT2AR表达的影响。方法:将SPF级雄性SD大鼠随机分为空白组和造模组,造模组用多平台水环境法制造慢性睡眠剥夺模型,造模成功后再将造模组大鼠随机分为模型组、西药组(舒乐安定)和酸枣仁汤组。用自主活动测试仪记录大鼠自主活动时间,用免疫组化和Western blot法检测大鼠海马中5-HT1AR和5-HT2AR的表达。结果:与空白组比较,模型组大鼠tL、tD、tall增加,大鼠活动时间增加(P<0.01);与模型组比较,西药组大鼠tL、tall和酸枣仁汤组大鼠tL、tD、tall减少,大鼠活动时间减少。空白组大鼠海马中5-HT1AR水平较高而5-HT2AR水平较低;造模后模型组大鼠海马中5-HT1AR水平下降而5-HT2AR水平上升;给药后西药组和酸枣仁汤组大鼠海马中5-HT1AR水平上升而5-HT2AR水平下降。结论:酸枣仁汤能调节大鼠海马中5-HT1AR和5-HT2AR的表达,从而改善睡眠剥夺大鼠睡眠状态,效果与舒乐安定相当。
Abstract:
Objective: To survey the effects of SuanZaoRen decoction on the expressions of 5-HT1AR and 5-HT2AR in hippocampus of the rats with chronic sleep deprivation(CSD). Methods: SPF male SD rats were randomized into the blank group and the modeling group, CSD rat models were prepared using modified multiple platform method(MMPM) in the modeling group, after successfully modeling, the rats in the modeling group were allocated to the model group, Western medicine(eurodin) group and SuanZaoRen decoction group. Autonomic activity tester was used to record autonomic activity time of the rats, immunohistochemical method and Western blot method were adopted to detect the expressions of 5-HT1AR and 5-HT2AR in hippocampus of the rats. Results: Compared with the blank group, tL, tD and tall of the rats in the model group increased, activity time added (P<0.01); compared with the model group, tL and tall of the rats in the Western medicine group, tL, tD and tall of the rats in SuanZaoRen decoction group decreased, activity time of the rats lowered. The levels of 5-HT1AR rose while the levels of 5-HT2AR lowered in the rats of the blank group; after modeling, the levels of 5-HT1AR decreased while the levels of 5-HT2AR rose in the rats of the model group; after medicating, the levels of 5-HT1AR rose while the levels of 5-HT2AR lowered in the rats of Western medicine group and SuanZaoRen decoction group. Conclusion: SuanZaoRen decoction could regulate the expressions of 5-HT1AR and 5-HT2AR in hippocampus of the rats, thus to improve the sleeping state of the rats equivalent to eurodin.
