目的:建立同时测定淫羊藿中木兰花碱、朝藿定C和淫羊藿苷含量的HPLC方法,研究淫羊藿(Epimedium brevicornu Maxim.)因产地不同带来的质量差异。方法:Venusil C18 MP色谱柱(250 mm×4.6 mm,5 μm),乙腈-1%KH2PO4梯度洗脱;流速1 mL/min。结果:木兰花碱、朝藿定C和淫羊藿苷分别在4.0~52.0,6.4~206.0,7.0~242.0 μg/mL范围内线性关系良好;回收率分别为97.66%、101.10%、96.76%,RSD分别为2.14%、2.20%、1.58%。结论:本方法适用于同时测定淫羊藿中生物碱和黄酮两种不同类型的成分;不同分布地区淫羊藿中朝藿定C、淫羊藿苷均呈现较大的变化,木兰花碱含量变化大,不适于作为质控成分。
Abstract:
Objective: To establish a HPLC method for determining the contents of magnoflorine, epimedin C and icariin in Yinyanghuo (epimedii folium) simultaneously, and study the quality difference caused by the producing areas. Methods: Venusil C18 MP chromatographic column(250 mm×4.6 mm, 5 μm), acetonitrile-1% KH2PO4 as gradient elution; flow rate was 1 mL/min. Results: Under the above chromatographic conditions, magnoflorine, epimedin C and icariin showed good linear relationship in the range between 4.0~52.0, 6.4~206.0 and 7.0~242.0 μg/mL; the recovery rates were 97.66%, 101.10% and 96.76%, RSD were 2.14%, 2.20% and 1.58% respectively. Conclusion: The method is suitable for simultaneous determination of alkaloid and flavone in Yinyanghuo; the contents of epimedin C and icariin show greater changes contained in Yinyanghuo in different distribution regions, the contents of magnoflorine vary, and it couldn't be taken as the ingredients for quality control.
