摘要:
目的:提高玫瑰花糖膏质量标准。方法:采用薄层色谱法对玫瑰花糖膏中的槲皮素、山柰酚、没食子酸3种成分进行定性鉴别;采用薄层色谱-生物自显影技术鉴别玫瑰花糖膏的抗氧化成分;依照药典测定玫瑰花糖膏中挥发性醚浸出物的含量和相对密度;根据玫瑰花糖膏中主要有效成分黄酮苷,大部分具有相同的苷元槲皮素和山柰酚的特点,加酸水解后采用HPLC法对总黄酮(以槲皮素、山柰酚之和计)进行含量测定,色谱条件:Symmetry C18色谱柱(4.6 mm×150 mm,5 μm),流动相:甲醇-0.4%磷酸(60∶40),进样量10 μL,流速:l.0 m L/min,柱温:30℃,检测波长:360 nm。结果:1)供试品(槲皮素、山柰酚、没食子酸)溶液在与对照品溶液色谱相应位置上,显相同颜色的斑点。2)玫瑰花糖膏中的没食子酸成分具有抗氧化活性。3)挥发性醚浸出物平均含量为0.1188%,相对密度平均为1.38。4)槲皮素在0.24~1.20 mg/mL范围内呈良好的线性关系,平均回收率(n= 6) 为100.43% ,RSD为1.52% ;山奈酚在0.08~0.40 mg/mL范围内呈良好的线性关系,平均回收率(n=6)为102.19%,RSD为1.80%。结论:本实验提高了玫瑰花糖膏的质量标准,为保证其临床应用的安全性和可控性奠定了基础。
Abstract:
Objective: To improve quality standard of rose massecuite. Methods: Quercetin, kaempferol and gallic acid contained in rose masseuite were qualitatively identified using TLC; antioxidant component in rose massecuite were identified by TLC-biological autoradiography technique; relative density and the contents of volatile ether extract in rose massecuite were measured according to pharmacopeia; the contents of total flavonoids (the summation of quercetin and kaempferol) were determined after acid hydrolysis according to main active ingredients flavonoid glycoside including the same glycoside quercetin and kaempferol. Chromatographic conditions: Symetry C18 chromatographic column (4.6 mm×150 mm, 5 μm), mobile phase: methanol-0.4% phosphoric acid (60:40), sample size 10μL, flow rate: 1.0mL/min, column temperature:30℃, detection wavelength: 360nm. Results: 1) The samples (quercetin, kaempferol and gallic acid) solution showed the same color spots in the corresponding sites of the color spectrum of the reference solution. 2)Gallic acid of rose massecuite possesses antioxidant activity. 3)Average contents of volatile ether extract were 0.1188%, average relative density was 1.38. 4)Quercetin showed the good linear relationship in the range between 0.24~1.20 mg/mL, average recovery rate (n=6) was 100.43%, RSD was 1.52%; kaempferol showed the good linear relationship in the range between 0.08~0.40 mg/mL, average recovery rate (n=6) was 102.19%, RSD was 1.80%. Conclusion: The experiment improve quality standard of rose massecuite, and it could lay the foundation for ensuring its safety and controllability in clinical application.
