摘要:
目的:建立关黄柏标准汤剂的质量控制方法。方法:收集具有代表性的10批关黄柏饮片,参照标准,制备标准汤剂。以盐酸巴马汀和盐酸小檗碱为定量检测指标,计算二者平均转移率与出膏率,并建立其HPLC指纹图谱分析方法。结果:对10批关黄柏饮片标准汤剂进行含量测定,指标成分的平均转移率为51.3%~65.9%,出膏率为17.3%~20.4%;并利用中药色谱指纹图谱相似度评价系统软件(2012版)进行分析,标定了10个共有峰,确认了其中3个峰,分别为盐酸药根碱、盐酸巴马汀、盐酸小檗碱;指纹图谱相似度均大于0.95。结论:建立了系统评价关黄柏饮片标准汤剂的质量评价方法,可为源于关黄柏水煎液的相关制剂的质量控制标准的制定提供参考。
Abstract:
Objective: To establish the methods for quality control of Guanhuangbo(phellodendri amurensis cortex) standard decoction. Methods: Ten batches of typical Guanhuangbo decoction pieces were collected and prepared into standard decoction in light of the preparation standard. Palmatine hydrochloric acid and berberine hydrochloride were chosen as the indexes of quantitative detection, to calculate average transfer rate and extraction rate, and establish HPLC fingerprint analysis methods for both indexes. Results: By determining the contents of ten batches of Guanghuangbo, average transfer rate of the index components was ranged from 51.3% to 65.9%, extraction rates was from 17.3% to 20.4%; the similarity evaluation system for herbal chromatographic fingerprint (edition 2012) was utilized to analyze, ten common peaks were determined, three were identified as root base hydrochloric acid, palmatine hydrochloric acid and berberine hydrochloride; the similarity of fingerprints were higher than 0.95. Conclusion: The quality evaluation method to assess Guanghuangbo standard decoction systematically is established, and it could provide the reference for establishing quality standard of Guanghuangbo related decoction.
