摘要:
目的:研究不同产地加工方法对党参饮片中党参炔苷、党参多糖、药典党参项下指标性成分的影响,并建立不同产地加工方法下党参指纹图谱,为党参不同产地加工方法质量控制及产地加工方法原理研究提供依据。方法:选用不同产地加工方式处理的党参样品,考察不同方法对党参饮片中党参炔苷含量,党参多糖含量等指标的影响,采用HPLC色谱法测定党参炔苷含量,采用用紫外-可见分光光度法测定党参多糖含量;采用HPLC-DAD检测法对10种不同产地加工方式处理的党参结,党参段进行指纹图谱研究,采用指纹图谱相似度软件进行分析。结果:党参炔苷进样量在0.33~6.55 μg范围内与峰面积呈良好的线性关系;党参多糖(以葡萄糖为对照)在0.294 8~2.528 2 mg/mL的范围内,浓度与吸光度值呈良好线性关系;以党参炔苷峰为参照峰,建立不同产地加工方法下党参HPLC-DAD指纹图谱,确认13个共有指纹峰,特征明显,结果可靠。党参经不同产地加工方法处理后品质亦有不同变化。结论:本实验所采用方法精密度、稳定性和重复性良好,符合含量测定及指纹图谱技术等要求,可作为合理选择党参产地加工方式的质量控制方法。
Abstract:
Objective: To provide the reference for the study on the principles and quality control for Dangshen processed by different methods from different producing areas by investigating the effects of the processing methods from different producing areas on Dangshen indicative components in pharmacopoeia, ginseng polysaccharide and codonopsis pilosula alkynyl glycoside contained in Dangshen, establishing the fingerprint of Dangshen processed by the methods from different producing areas. Methods: After processing the samples with different methods from different producing areas, the effects of different processing methods were surveyed on the contents of ginseng polysaccharide and codonopsis pilosula alkynyl glycoside contained in Dangshen, HPLC chromatographic method was adopted to determine the contents of codonopsis pilosula alkynyl glycoside, CAPCELL PAK C18-MG Ⅱ column (4.6 mm×250 mm, 5 μm), mobile phase was methanol: water (28:72), flow rate 1.0mL/min, column temperature 30℃, detection wavelength 267nm; ultraviolet-visible spectrophotometry was used to measure the contents of codonopsis pilosula polysaccharide; HPLC-DAD detection method was employed to study the fingerprint print of the knot and part of Dangshen processed by ten methods from different producing areas, and the fingerprint was analyzed using fingerprint similarity software. Results: Sample size of codonopsis pilosula alkynyl glycoside showed better linear relationship in the rang from 0.33 to 6.55 μg and peak area; when codonopsis pilosula polysaccharide (glucose as the control) was ranged from 0.294 8 to 2.528 2 mg/mL, the concentrations and absorbance value were in good linear relationship; by taking the peak of codonopsis pilosula alkynyl glycoside as the control, HPLC-DAD fingerprint of Dangshen was established after processed with different methods, finally 13 common fingerprint peaks were confirmed, and they presented the obvious characteristics and reliable results. The property of Dangshen has changed differently after processed with different methods of different producing areas. Conclusion: The method employed in the experiment shows better precision, stability and repeatability, meets the requirements of content determination and fingerprint technique, and it could be taken as the method of quality control for rationally choosing processing methods of Dangshen.
