摘要:
目的:建立玄参中哈巴俄苷、肉桂酸、毛蕊花糖苷及安格洛苷C含量快速测定近红外光谱定量的模型。方法:以高效液相色谱法(HPLC)检测值作为参照,采用傅里叶变换近红外漫反射光谱技术采集玄参的近红外光谱,结合偏最小二乘法(PLS)分别建立哈巴俄苷、肉桂酸、毛蕊花糖苷及安格洛苷C含量的快速测定方法。结果:玄参中哈巴俄苷、肉桂酸、毛蕊花糖苷及安格洛苷C的校正模型相关系数(R)、校正均方差(root mean square error of calibration,RMSEC)、预测均方差(root mean square error of prediction,RMSEP)分别为0.967 05、0.0101、0.025 32;0.961 76、0.0141、0.024 43;0.998 53、0.008 89、0.027 32;0.961 34、0.0189、0.021 91。通过外部验证,玄参中四种成分的预测值与真实值相关系数分别达为0.984 2(哈巴俄苷)、0.9960(肉桂酸)、0.9994(毛蕊花糖苷)、0.9939(安格洛苷)。结论:该方法准确、快速、简便,可实现大批量样品的快速分析。
Abstract:
Objective: To establish the calibration model of near infrared spectroscopy (NIRS) for rapid determination of harpagoside, cinnamic acid, acteoside and angoroside C contained in Xuanshen (Scrophulariae radix). Methods: By taking HPLC detection value as the reference, Fourier transform near infrared diffuse reflectance spectroscopy was used to collect NIRS of Xuanshen, the methods for rapid determination of harpagoside, cinnamic acid, acteoside and angoroside C were established respectively based on partial least square method (PLS). Results: Corrected model related coefficients(R), RMSEC and RMSEP of harpagoside, cinnamic acid, acteoside and angoroside C contained in Xuanshen were 0.967 05, 0.0101 and 0.025 32; 0.961 76, 0.0141 and 0.024 43; 0.998 53, 0.008 89 and 0.027 32; 0.961 34, 0.0189 and 0.021 91. Correlation coefficients of the predicted values and true values of four ingredients in Xuanshen reached 0.9842 (harpagoside), 0.9960 (cinnamic acid), 0.9994(acteoside) and 0.9939 (angoroside C) through external validation. Conclusion:The method, accurate, rapid and simple, could realize rapid analysis of the samples in bulk.
