摘要:
目的:探讨绿原酸对结肠癌细胞HCT116增殖和凋亡的影响及其可能的作用机制。方法:采用CCK8实验方法检测绿原酸对HCT116细胞增殖的影响,流式细胞术检测绿原酸对HCT116细胞凋亡的影响,免疫印迹技术检测绿原酸对HCT116细胞中磷脂酰肌醇3-激酶(phosphatidylinositol 3-kinase,PI3K)、p-PI3K、蛋白激酶B(protein kinase B,Akt)、p-Akt、B淋巴细胞瘤(B-cell lymphoma-2,Bcl-2)及Bax蛋白表达的影响。结果:与空白对照组相比,绿原酸能明显抑制HCT116细胞增殖,差异具有统计学意义(P<0.05);绿原酸能显著诱导HCT116细胞凋亡,差异具有统计学意义(P<0.05);绿原酸能明显抑制p-PI3K、p-Akt、Bcl-2蛋白表达,诱导Bax蛋白表达,且呈浓度依赖性。结论:绿原酸能明显抑制HCT116细胞的增殖、诱导其凋亡,其作用可能与抑制PI3K/Akt信号通路活化有关。
Abstract:
Objective: To study the effects of chlorogenic acid on proliferation and apoptosis of colon cancer cells and its possible mechanism. Methods: CCK8 assay was carried out to investigate the effect of chlorogenic acid on the proliferation of HCT116 cells, flow cytometry assay to assess the effects of chlorogenic acid on the apoptosis of HCT116 cells, Western blot to determine the expressions of PI3K, p-PI3K, Akt, p-Akt, Bcl-2 and Bax in HCT116 cells. Results: Compared with blank control group, chlorogenic acid could inhibit the proliferation of HCT116 cells obviously, and the difference showed statistical meaning (P<0.05); it could notably induce the apoptosis of HCT116 cells, and the difference had statistical meaning (P<0.05); it could inhibit the expressions of p-PI3K, p-Akt, Bcl-2 protein, induce the expressions of Bax protein, in a dose-dependent manner. Conclusion: Chlorogenic acid could inhibit the proliferation of HCT116 cells, induce its apoptosis, and its mechanism might be related to inhibiting the activation of PI3K/Akt signaling pathway.
