摘要:
目的:探讨藏茵陈对重症胰腺炎大鼠TLR9/MyD88/NF-κBp65信号通路的影响。方法:将大鼠分为假手术组、模型组及藏茵陈高、中、低剂量组,连续灌胃给药14天后造模。采用酶联免疫吸附法测定各组大鼠白细胞介素1(interleukin-1,IL-1)、白细胞介素6(interleukin-6,IL-6)、肿瘤坏死因子α(tumor necrosis factor α,TNF-α)及Toll样受体(toll-like receptors 9,TLR9)、髓样分化因子(Myeloid differentiation primary response gene,MyD88)、核转录因子kappaB p65(nuclear factor kappaBp65,NF-κBp65)的蛋白表达;实时荧光定量PCR测定IL-1、IL-6、TNF-α及TLR9、MyD88、NF-κBp65的基因表达。结果:与假手术组比较,模型组大鼠IL-1、IL-6、TNF-α及TLR9、MyD88、NF-κBp65的蛋白和基因表达均升高(P<0.05);与模型组比较,藏茵陈高剂量组能够降低大鼠胰腺组织IL-1、IL-6、TNF-α及TLR9、MyD88、NF-κBp65的蛋白和基因表达(P<0.05)。结论:藏茵陈能阻断TLR9/MyD88/NF-κBp65信号通路,减轻重症胰腺炎大鼠炎症反应。
Abstract:
Objective: To study the effects of Tibetan medicine Yinchen(swertia mussotii Franch) on TLR9/MyD88/NF-κBp65 signaling pathway in the rats with severe acute pancreatitis(SAP). Methods: All the rats were allocated to sham operation group, the model group, high, moderate and low dose groups of Tibetan medicine Yinchen, the models were successfully prepared by intragastric administration for 14 days consecutively. ELISA method was used to detect the levels of IL-1, IL-6 and TNF-α, and the expressions of TLR9, MyD88 and NF-κBp65 proteins; real-time fluorescent quantitative PCR to measure the expressions of IL-1, IL-6 and TNF-α, and the expressions of TLR9, MyD88 and NF-κBp65 proteins. Results: compared with shan operation group, the expressions of IL-1, IL-6 and TNF-α, TLR9, MyD88 and NF-κBp65 genes and proteins rose (P<0.05); compared with the model group, high dose group of Tibetan medicine Yinchen could reduce the expressions of IL-1, IL-6 and TNF-α, TLR9, MyD88 and NF-κBp65 genes and proteins(P<0.05). Conclusion: Tibetan medicine Yinchen could block TLR9/MyD88/NF-κBp65 signaling pathway, and relieve inflammatory reaction of SAP rats.
