摘要:
目的:优选加味黄连解毒合剂提取工艺的最佳条件并建立质量标准。方法:以黄芩苷含量、干膏率和澄明度为评价指标,用单因素考察方法筛选合剂纯化工艺;以干膏率、黄芩苷含量为评价指标,采用正交实验法筛选黄连解毒合剂的制备工艺;采用薄层色谱法对黄连、栀子、金银花进行薄层定性鉴别,采用高效液相色谱法测定黄芩中黄芩苷的含量。结果:最佳提取工艺为煎煮2次,每次加水12倍,每次煎煮1 h,滤过,滤液75℃下离心半径8 cm,4000 r/min离心20 min。薄层色谱鉴别斑点显色清晰,阴性无干扰,以高效液相色谱法所测定黄芩苷含量在1.006~5.030 μg范围内呈良好线性关系(r=0.9996,n=5),平均回收率为98.39%。结论:优选的工艺合理可行,提取效率高,所建立的质量控制方法专属性强,重复性好,可用于加味黄连解毒合剂的质量控制。
Abstract:
Objective: To optimize the conditions for extraction process of modified Huanglian Jiedu mixture and establish the quality standard. Methods: Single factor survey method was used to screen the purification technology of the mixture by taking the contents, extraction rate and clarity of baicalin as the assessment indexes; orthogonal test was adopted to optimize the preparation technology of Huanglian Jiedu mixture by taking the the contents and extraction rate of baicalin as the assessment indexes; TLC was used to perform qualitative identification of Huanglian (Coptis chinensis Franch.), Zhizi (Gardenia jasminoides Ellis) and Jinyinhua (Lonicera Japonica), HPLC to detect the contents of baicalin in Huangqin. Results: The best extraction process was decocting twice, adding water 12 times each time, decocting for one hour each time, filtration, the filtrate was centrifuged for 20 min urder 75℃ with the radius 8 cm, 4000 r/min. TLC identification showed that the spots were clear, negative without interference, HPLC resultspresented that baicalin displayed better linear relationship in the range between 1.006 and 5.030 μg(r=0.9996, n=5), average recovery rate was 98.39%. Conclusion: The optimized technology is rational and feasible with high extraction efficiency, the method, specific and reproducible, could be used for quality control of modified Huanglian Jiedu mixture.
