摘要:
目的研究龙葵总碱对骨髓瘤移植裸鼠磷酸化核因子-κB抑制蛋白(inhibitor kappa b alpha,IκB-α)、核因子-κB(nuclear factor κB,NF-κB)/干扰素调节因子4(interferon regulatory factor 4,IRF4)信号通路表达的影响,明确龙葵总碱抗骨髓瘤的作用靶点和机制。 方法传代培养人骨髓瘤细胞RPMI8226,选取90只裸鼠,随机分为空白组,模型组,硼替佐米组,龙葵总碱低、中、高剂量组,每组15只。除空白组外,其余小鼠制备多发性骨髓瘤模型,造模成功后连续灌胃给药龙葵总碱14天,皮下注射硼替佐米14天,断颈处死裸鼠,取出瘤体组织。瘤组织细胞应用CCK8检测不同浓度的龙葵总碱和硼替佐米对骨髓瘤细胞增殖的影响,采用流式细胞仪检测瘤组织细胞凋亡率;采用Western blot检测IκB-α、磷酸化IκB-α、NF-κB、P65、IRF4表达。 结果龙葵总碱能明显抑制人骨髓瘤细胞的增殖,呈时间和剂量依赖性,并能促进骨髓瘤组织细胞的凋亡,呈现剂量依赖性。Western blot显示龙葵总碱高、中剂量组磷酸化IκB-α、NF-κB与IRF4表达均显著减少,而其负调控因子IκB-α水平明显升高。 结论龙葵总碱通过抑制磷酸化IκB-α,引起NF-κB失活,促进骨髓瘤细胞IRF4表达减少,在一定程度上促使骨髓瘤细胞凋亡。
Abstract:
ObjectiveTo study the effects of total alkaloids of solanum nigrum on the expressions of phosphorylated IκB-α and NF-κB/IRF4 signaling pathway in transported-myeloma in nude mice, and survey anti-myeloma targets and mechanism of solanum nigrum. MethodsHuman myeloma cell line RPMI8226 was cultured for serial passage, 90 naked mice were chosen and randomized into the blank group, the model group, bortezomib group, low, moderate and high dosages groups of total alkaloids of solanum nigrum, 15 mice each group. Except the blank group, the rest were established into mice models with multiple myeloma, after successful modeling, they were drenched with total alkaloids of solanum nigrum for 14 days consecutively, after they accepted subcutaneous injection of bortezomib for 14 days, the naked mice were sacrificed by cervical dislocation to get out the tumor tissues. CCK8 method was adopted to detect the effects of different concentrations of total alkaloids of solanum nigrum and bortezomib on the proliferation of myeloma cells, flow cytometry was used to detect the apoptosis rate of tumor tissue; Western blot was used to detect the expressions of IκB-α, phosphorylated IκB-α, NF-κB, p65 and IRF4. ResultsTotal alkaloids of solanum nigrum could notably inhibit the proliferation of human myeloma cells, in time- and dose-dependent manner, it could promote the apoptosis of myeloma cells, in the dose-dependent manner. Western blot displayed that the expressions of p-IκB-α, NF-κB and IRF4 lowered in moderate and high doses groups of total alkaloids of solanum nigrum, while the levels of negative regulatory factor IκB-α rose apparently. ConclusionTotal alkaloids of solanum nigrum could promote the apoptosis of myeloma cells to a certain degree through inhibiting the expressions of phosphorylated IκB-α, inducing the inactivation of NF-κB and reducing the expressions of IRF4 in human myeloma cells.
