摘要:
目的观察刺山柑对佐剂型关节炎大鼠血清白细胞介素17(interleukin-17,IL-17)、IL-6、IL-10和IL-23及转化生长因子β(transforming growth factor-β,TGF-β)含量的影响,探讨其对免疫Th17/Treg平衡调节的作用机制。 方法将40只Wistar大鼠随机分为正常组、模型组、阳性药组和刺山柑组各10只。于大鼠足跖皮下注射0.1 mL弗氏完全佐剂建立类风湿性关节炎(rheumatoid arthritis,RA)模型。造模成功后3天,阳性药组灌胃甲氨蝶呤0.1 mg/100 g,刺山柑组外敷刺山柑于大鼠左后足踝,各组均治疗3个疗程(21天)。分别在每个疗程结束后观察大鼠AI指数评分和足爪容积,并用ELISA检测大鼠血清IL-17、IL-6、IL-10、IL-23及TGF-β含量。 结果造模后大鼠关节肿胀明显,血清IL-17、IL-6、IL-23升高,TGF-β降低,差异具有统计学意义(P<0.O1)。治疗后阳性药组和刺山柑组IL-17含量均降低(P<0.01),刺山柑组IL-6降低、IL-10升高(P<0.05)。 结论刺山柑对RA的消炎作用可能是通过调节Th17和Treg细胞水平,改善Th17/Treg失衡状态实现。
Abstract:
ObjectiveTo observe the effects of Capparis spinosa on the levels of IL-17, IL-6, IL-10, IL-23 and TGF-β in the serum of rats with adjuvant arthritis, to investigate its mechanism of regulating Th17/Treg immune balance. MethodsForty rats were randomized into the normal group, the model group, positive medicine group and Capparis spinosa group. RA rat models were established by subcutaneous injection of 0.1 mL Freund's complete adjuvant. Positive medicine group were drenched with methotrexate, 0.1 mg/100 g and Capparis spinosa group accepted external application of Capparis spinosa over left posterior ankles three days after successful modeling, different groups were treated for three courses (21 days). By the end of each course, to observe AI index score and paw volume of the rats respectively, ELISA method was used to measure the contents of IL-17, IL-6, IL-10, IL-23 and TGF-β in the serum of rats. ResultsAfter modeling, joint swelling was notable, the levels of IL-17, IL-6 and IL-23 in the serum rose, the levels of TGF-β lowered, and the difference was statistically significant (P<0.01). After treating, the contents of IL-17 decreased in positive medicine group and Capparis spinosa group (P<0.01), the levels of IL-6 lowered and the levels of IL-10 rose in Capparis spinosa group (P<0.05). ConclusionAnti-inflammatory effect of Capparis spinosa on RA could be realized through regulating Th17 and Treg cellular levels and improving Th17/Treg imbalance.
