摘要:
目的探讨杨梅素对心肌缺血再灌注损伤(myocardial ischemia reperfusion injury,MI/RI)大鼠的保护作用及对血红素加氧酶1(heme oxygenase-1,HO-1)的影响。 方法选择雄性SD大鼠72只按照随机数字法分为假手术组,模型组,阳性对照组,杨梅素高、中、低剂量组,每组12只。相应药物干预2周后构建MI/RI模型(缺血30 min,再灌注120 min),收集心脏标本和血液,HE染色检测心肌病理学,全自动生化分析仪检测血清乳酸脱氢酶(lactate dehydrogenase,LDH)、肌酸激酶(creatine kinase,CK),ELISA法检测心肌超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、白细胞介素6(interleukin-6,IL-6)和肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)水平,采用Western Blotting和QT-PCR检测心肌HO-1蛋白及mRNA表达特点。 结果模型组有明显心肌纤维损伤,而杨梅素各剂量组对其则有一定的调节作用,其中杨梅素高剂量组心肌细胞仅有少量变性。与假手术组相比,模型组大鼠血清LDH和CK明显升高(P<0.05),心肌MDA、IL-6和TNF-α明显升高(P<0.05),心肌SOD明显下降(P<0.05);与模型组相比,杨梅素各剂量组和阳性对照组LDH、CK、MDA、IL-6和TNF-α均下降(P<0.05),SOD升高(P<0.05),其中以杨梅素高剂量组最显著。与假手术组相比,模型组HO-1蛋白及mRNA表达明显降低(P<0.05),杨梅素各剂量组和阳性对照组HO-1蛋白及mRNA表达则明显升高(P<0.05),其中以杨梅素高剂量组改善最显著。 结论杨梅素预防给药可明显抑制MI/RI大鼠心肌损伤,其作用机理可能与杨梅素可升高HO-1表达,进而改善炎症刺激同时提高抗氧化能力有关。
Abstract:
ObjectiveTo explore protective effects of myricetin on myocardial ischemia reperfusion injury (MI/RI) in rats and its influence on HO-1. MethodsAll 72 male SD rats were chosen and allocated to sham operation group, the model group, positive control group, high, moderate and low doses groups of myricetin according to random number method, 12 rats in each group. After two weeks of drug intervention, MI/RI models were established (ischemia for 30 minutes, reperfusion for 120 minutes) to collect the samples of heart and blood, HE staining was used to detect myocardial pathology, automatic biochemical analyzer to measure the levels of LDH and CK, ELISA method to measure the levels of SOD, MDA, IL-6 and TNF-α, Western blotting and QT-PCR were utilized to measure the expressions of HO-1 protein and mRNA. ResultsThe model group had obvious myocardial fiber injury, while different doses groups of myricetin had a certain regulatory effect on it, among them, there was only a little degeneration of cardiomyocytes in high dose group of myricetin. Compared with sham operation group, the levels of LDH and CK rose in the model group (P<0.05), the levels of myocardial MDA, IL-6 and TNF-α increased notably (P<0.05), myocardial SOD decreased obviously (P<0.05); compared with the model group, the levels of LDH, CK , MDA, IL-6 and TNF-α dropped in different dose groups of myricetin and positive control group(P<0.05), myocardial SOD rose (P<0.05), among them, different indexes were the most significant in high dose group of myricetin. Compared with sham operation group, the expressions of myocardial HO-1 protein and mRNA decreased notably in the model group (P<0.05), the expressions of myocardial HO-1 protein and mRNA went up notably in different dose groups of myricetin and positive control group(P<0.05), among them, all the indexes were significant in high dose group of myricetin. ConclusionPreventive administration of myricetin could notably inhibit myocardial damage, and its mechanism might be related to myricetin increasing HO-1 expression, improving inflammation and antioxidant capacity.
