摘要:
目的基于网络药理学探讨葶苈大枣泻肺汤治疗慢性心力衰竭(chronic heart failure,CHF)的机制。 方法利用中药系统药理学分析平台(traditional Chinese medicine systems pharmacology,TCMSP)获取葶苈子、大枣的活性成分,采用SwissTargetPrediction、GeneCards数据库、STRING数据库、DAVID 6.8数据库及软件筛选分析治疗CHF的作用靶点,进行基因本体论(gene ontology,GO)分类富集分析和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)通路富集分析。 结果筛选出葶苈子、大枣活性成分共42个,作用于498个靶点,主要通过PI3K/Akt信号通路、cAMP信号通路、钙离子信号通路、胰岛素抵抗、甲状腺激素信号通路、T细胞受体信号通路、HIF-1等信号通路作用于CHF。 结论葶苈大枣泻肺汤可通过多成分、多靶点、多通路治疗慢性心力衰竭。
Abstract:
ObjectiveTo discuss the mechanism of Tingli Dazao Xiefei Tang in treating CHF based on network pharmacology. MethodsActive ingredients of Tinglizi and Chinese date were gained using TCMSP, targets of treating CHF were selected and analyzed using SwissTargetPrediction, GeneCards database, STRING database, DAVID 6.8 database and software, to carry out classification and enrichment analysis of GO classification and enrichment analysis, and KEGG pathway enrichment analysis. ResultsA total of 42 active components were screened out from Tinglizi and Chinese date, which acted on 498 targets, and they mainly acted on CHF through phosphatidylinositol 3-kinase/serine threonine kinase (PI3K/Akt) signaling pathway, cAMP signaling pathway, calcium signaling pathway, insulin resistance, thyroid hormone signaling pathway, T cell receptor signaling pathway, hypoxia inducible factor-1 (HIF-1) and others. ConclusionTingli Dazao Xiefei Tang could treat CHF by multi-component, multi-target and multi-channel therapy.
