摘要:
目的探讨小柴胡汤化裁方对阿尔茨海默病(Alzheimer’s disease,AD)小鼠记忆认知能力及海马形态学的影响。 方法选取SPF级小鼠60只,随机分成6组,除正常组外,其余各组应用脑立体定位仪定位海马组织并注射β淀粉样蛋白1-42(amyloid β-protein 1-42Aβ1-42)造模,中药组灌胃小柴胡汤化裁方高、中、低剂量(40、20、10 g/kg),西药组灌胃尼莫地平(15 mg/kg)。用Morris水迷宫观察AD小鼠定位航行和空间探索行为变化情况,同时观察AD小鼠海马神经元形态。 结果模型组AD小鼠的潜伏期增加,跨越平台次数减少,中药尤其是高剂量干预的AD小鼠潜伏期缩短,跨越平台次数增加。与模型组比较,中药高、中剂量组和西药组小鼠海马神经细胞形态及神经元丧失减轻。 结论小柴胡汤化裁方能有效改善AD小鼠学习记忆能力,保护海马神经元组织。
Abstract:
ObjectiveTo explore the effects of modified Xiaochaihu Tang on memory and cognitive ability and the morphology of hippocampal neurons in AD mice. MethodsSixty SPF mice were chosen and randomly allocated to six groups, except the normal group, the mice in other groups were prepared into the models by injecting amyloid beta protein (A β1-42) into hippocampal tissue located by stereotactic apparatus, the mice of herbal group were drenched with high, moderate and low doses of modified Xiaochaihu Tang (40, 20 and 10 g/kg), and Western medicine group with nimodipine (15 mg/kg). Morris water maze was used to observe the changes in positioning navigation and space exploration behavior of AD mice, simultaneously, to observe the morphology of hippocampal neurons in AD mice. ResultsIn the model group, the latency of AD mice increased and the number of crossing platform decreased, the latency of AD mice with high dose of herbs intervention was reduced, and the number of crossing platforms increased. Compared with the model group, the morphology and loss of neurons in hippocampus of the high and moderate dose groups and the Western medicine group were reduced. ConclusionModified Xiaochaihu Tang could effectively improve memory and cognitive ability, and protect hippocampal neurons in AD mice.
