摘要:
目的探讨当归多糖对慢性应激抑郁小鼠的影响及其作用机制。 方法将60只C57BL/6小鼠随机分为正常对照组、模型对照组、当归多糖低剂量组(20 mg/kg当归多糖灌胃)、当归多糖高剂量组(40 mg/kg当归多糖灌胃)及氟西汀组(10 mg/kg氟西汀灌胃),每组12只。使用慢性应激方法诱导小鼠抑郁行为并在造模的同时灌胃给药。使用行为学评估方法评价慢性应激小鼠抑郁行为[糖水偏好率、强迫游泳测试(forced swimming test,FST)、悬尾测试(tail suspension test,TST)和高架十字迷宫实验(elevated plus maze test,EPMT)];使用LC-MS/MS色谱法检测小鼠海马组织中神经递质含量[多巴胺(Dopamine,DA)、5羟色胺(5-hydroxytryptamine,5-HT)、γ-氨基丁酸(γ-aminobutyric acid,GABA)、血糖(Glucose,GLU)和GABA/GLU率];使用qPCR法检测小鼠海马组织中色氨酸羟化酶1(recombinant tryptophan hydroxylase 1,TPH1 mRNA)和人突触小体相关蛋白(synaptosomal associated protein 25,SNAP25 mRNA)表达。 结果40 mg/kg当归多糖能够明显增加小鼠FST与TST累积不动时间(P<0.05),降低小鼠EMPT中进入开臂时间及糖水偏好率(P<0.05),减少鼠海马组织中DA、5-HT含量、TPH1 mRNA表达量及GABA/GLU比率(P<0.05)。 结论当归多糖能够改善慢性应激模型小鼠抑郁行为,其作用机制与上调TPH1 mRNA表达,引起5-HT合成增加,提高DA含量及升高GABA/GLU比率有关,可以调节神经递质传导,纠正小鼠兴奋抑制功能紊乱,但与调节SNAP25 mRNA表达变化无关。
Abstract:
ObjectiveTo discuss the influence of angelica polysaccharide on the behavior of chronic stress depression mice and its mechanism. MethodsSixty C57BL/6 mice were randomly allocated to normal control group, model control group, low dose group of angelica polysaccharide (20 mg/kg angelica polysaccharide by gavage), high dose group of angelica polysaccharide(40 mg/kg angelica polysaccharide by gavage) and fluoxetine group (10 mg/kg fluoxetine by gavage), 12 mice in each group. Chronic stress was used to induce depressive behavior in mice, simultaneously, the drug was given by gavage.Behavioral assessment method was adopted to evaluate depressive behavior of chronic stress mice(sugar preference rate, FST, TST and EPMT); LC-MS/MS chromatography was used to test the contents of neurotransmitter(DA, 5-HT, GABA, GLU and GABA/GLU rate) in hippocampal tissue of mice;qPCR method was used to detect the expressions of TPH1mRNA and SNAP25mRNA in hippocampal tissue of mice. ResultsThe results found that 40mg/kg angelica polysaccharide could notably increase cumulative immobility time of FST and TST in mice(P<0.05), reduce the time to open arm of EMPT and sugar preference rate(P<0.05), lower the contents of DA and 5-HT in hippocampal tissue of mice, the expressions of TPH1 mRNA and GABA/GLU rate(P<0.05). ConclusionAngelica polysaccharide could improve depressive behavior in chronic stress mice model,and its mechanism is related to upregulating the expressions of TPH1mRNA,increasing the synthesis of 5-HT, lifting the contents of DA and raising GABA/GLU rate, and then to regulate the transmission of neurotransmitter,correct the disturbance of excitatory inhibition in mice, however, it is not related to the regulation of SNAP25mRNA expression.
