摘要:
目的研究人参皂苷Rb1对兔离体肠平滑肌收缩活动的影响。 方法采用兔离体肠平滑肌实验,应用BL-420F生物机能实验系统,观察人参皂苷Rb1对正常状态下兔离体肠平滑肌自发性收缩的影响,探究人参皂苷Rb1影响兔离体肠平滑肌收缩的作用机制。 结果人参皂苷Rb1能浓度依赖性地抑制兔离体肠自发性收缩,且浓度在70、90 μmol/L可明显降低兔离体小肠平滑肌自发性收缩张力和收缩振幅(P<0.01),但对频率无影响(P>0.05)。用钌红、肝素和维拉帕米对兔离体肠平滑肌预处理后,人参皂苷Rb1对兔离体肠平滑肌收缩的抑制程度明显增强(P<0.05)。 结论人参皂苷Rb1可显著抑制兔离体肠平滑肌自发性收缩,其机制可能是阻断L-型Ca2+通道抑制外钙内流,或阻断ryanodine受体和三磷酸肌醇(inositol triphosphate,IP3)受体抑制内钙释放,最终使细胞内钙离子浓度降低,从而抑制肠平滑肌的收缩。
Abstract:
ObjectiveTo study the effects of ginsenoside Rb1 on the contraction of isolated rabbit intestine smooth muscle. MethodsThe experimental study on the isolated rabbit intestinal smooth muscle, and BL-420F biological function experiment system were applied to observe the effects of ginsenoside Rb1 on spontaneous contraction of the isolated rabbit intestinal smooth muscle under normal state, and explore the mechanism of ginsenoside Rb1 acting on the contraction of isolated rabbit intestine smooth muscle. ResultsGinsenoside Rb1 could inhibit spontaneous contraction of the isolated rabbit intestinal smooth muscle in dose-dependent manner, it could reduce spontaneous contractile tension and amplitude of isolated small intestinal smooth muscle in rabbits in the concentrations of 70 and 90 μmol/L notably (P<0.01), while it showed no effects on the frequency (P>0.05). After preconditioning the isolated rabbit intestinal smooth muscle with ruthenium red,heparin and verapamil, the inhibitory degrees of ginsenoside Rb1 enhanced notably on the contraction of isolated rabbit intestine smooth muscle (P<0.05). ConclusionGinsenoside Rb1 could significantly inhibit spontaneous contraction of the isolated rabbit intestinal smooth muscle, and its mechanism may be inhibiting calcium influx by blocking L-type Ca2+ channel, or inhibiting the release of intracellular calcium by blocking ryanodine receptor and IP3 receptor, eventually reduces the concentration of intracellular calcium, thereby inhibiting the contraction of intestinal smooth muscle.
