摘要:
目的探讨高血压病气虚血瘀证患者血清对人脐静脉内皮细胞株(human umbilical vein endothelial cell line 1730,CRL-1730)细胞色素C(cytochrome-c,Cyt-C)凋亡途径的影响,以探讨其诱导内皮细胞凋亡的机制。 方法以高血压病气虚血瘀证患者血清作用于体外培养的CRL-1730,激光共聚焦显微镜、蛋白免疫印迹(Western Blot)和酶联免疫法(ELISA)检测内皮细胞Cyt-C、Apaf-1、Caspase-9蛋白的表达、分布和含量。 结果激光共聚焦显微镜结果显示,与健康对照组比较,高血压病气虚血瘀证组及高血压病非气虚血瘀证组Cyt-C的表达均明显升高(P<0.01);与高血压病气虚血瘀证组比较,补阳还五汤各剂量组Cyt-C的表达明显降低(P<0.05)。Western Blot结果显示:与健康组比较,高血压病气虚血瘀证组及高血压病非气虚血瘀证组Cyt-C、Apaf-1、Caspase-9的表达均明显升高(P<0.05);与高血压病气虚血瘀证组比较,补阳还五汤各剂量组Cyt-C、Apaf-1、Caspase-9的表达明显下降(P<0.05)。ELISA结果显示:与健康组比较,高血压病气虚血瘀证组及高血压病非气虚血瘀证组Cyt-C、Apaf-1、Caspase-9含量均明显升高(P<0.05);与高血压病非气虚血瘀证组比较,高血压病气虚血瘀证组Cyt-C、Apaf-1、Caspase-9含量明显升高(P<0.05);与高血压病气虚血瘀证组比较,补阳还五汤各剂量组Cyt-C、Apaf-1、Caspase-9含量明显降低(P<0.05)。 结论高血压病气虚血瘀证患者血清可诱导CRL-1730凋亡,其机制可能与线粒体Cyt-C途径相关蛋白的表达升高有关,补阳还五汤能减轻凋亡。
Abstract:
ObjectiveTo survey the mechanism of the apoptosis of endothelial cells induced by the serum by exploring the serum in patients with hypertension of Qi deficiency and blood stasis pattern on CRL-1730 Cyt-C apoptosis pathway. MethodsCRL-1730 cultured in vitro was dealt with the serum of the patients with Qi deficiency and blood stasis pattern hypertension, laser confocal microscope, Western Blot and ELISA were used to detect the expressions, distribution and contents of Cyt-C, Apaf-1 and Caspase-9 protein in endothelial cells. ResultsThe results of laser confocal microscope displayed: compared with healthy control group, the expressions of Cyt-C were increased notably in the group of Qi deficiency and blood stasis pattern hyptertension and the group of non Qi deficiency and blood stasis pattern hyptertension (P<0.01); compared with the group of Qi deficiency and blood stasis pattern hyptertension, the expressions of Cyt-C were lowered remarkably in different dosages groups of Buyang Huanwu decoction (P<0.05). The results of Western Blot showed: compared with healthy group, the expressions of Cyt-C, Apaf-1 and Caspase-9 were significantly increased in the group of Qi deficiency and blood stasis pattern hyptertension and the group of non Qi deficiency and blood stasis pattern hyptertension (P<0.05); compared with the group of Qi deficiency and blood stasis pattern hyptertension, the expressions of Cyt-C, Apaf-1 and Caspase-9 significantly decreased in different dosages groups of Buyang Huanwu decoction (P<0.05). ELISA results showed: compared with healthy group, the contents of Cyt-C, Apaf-1 and Caspase-9 were notably raised in the group of Qi deficiency and blood stasis pattern hyptertension and the group of non Qi deficiency and blood stasis pattern hyptertension (P<0.05); compared with the group of non Qi deficiency and blood stasis pattern hyptertension, the contents of Cyt-C, Apaf-1 and Caspase-9 were apparently elevated in the group of Qi deficiency and blood stasis pattern hyptertension (P<0.05); compared with the group of Qi deficiency and blood stasis pattern hyptertension, the contents of Cyt-C, Apaf-1 and Caspase-9 were decreased notably in different dosages groups of Buyang Huanwu decoction (P<0.05). ConclusionThe serum of the patients with Qi deficiency and blood stasis pattern hyptertension could induce the apoptosis of CRL-1730, and its mechanism might be related to the increa-sed expressions of mitochondrial Cyt-C pathway related proteins, and Buyang Huanwu decoction could relieve the apoptosis.
