摘要:
目的探讨芪葵颗粒干预2型糖尿病合并骨质疏松的作用机制。 方法将18只雌性SD大鼠随机分为假造模组、模型对照组及治疗组,每组6只。使用卵巢切除术+链尿佐菌素(streptozotocin,STZ)的方法进行造模。治疗组造模后每日予芪葵颗粒灌胃,假造模组、模型对照组给予等量生理盐水干预。干预3周后,测定各组大鼠血糖(glucose,Glu)、甘油三酯(triglyceride,TG)、体质量(body weight,BW)、骨密度(bone mineral density,BMD),并取各组大鼠间充质干细胞(mesenchymal stem cells,MSCs)进行体外培养,通过MTT测定MSCs增殖活性,使用茜素红染色及ELISA对骨碱性磷酸酶(bone-alkaline phosphatase,B-ALP)和骨钙素(osteocalcin,BGP)定量测定MSCs成骨分化能力,油红O染色测定MSCs成脂肪分化能力。 结果给药3周后,与模型对照组比较,治疗组大鼠的Glu降低、骨密度增高(P<0.05);模型对照组矿化钙结节数目较少(P<0.05);模型对照组脂肪细胞数显著多于其他两组(P<0.05);各组BMCSs成骨诱导后,模型对照组BGP及B-ALP含量均显著低于假造模组(P<0.05),而芪葵颗粒组BGP及B-ALP水平升高(P<0.05),且MTT提示细胞增殖活性提高(P<0.05)。 结论芪葵颗粒能在改善血糖的基础上提高大鼠模型骨密度,体外细胞实验表明芪葵颗粒增加了模型大鼠MSCs的增殖活性,提高其成骨分化能力,同时下调了MSCs向脂肪分化的能力。
Abstract:
ObjectiveTo investigate the mechanism of Qikui granules in the intervention for type 2 diabetes rats with osteoporosis. MethodsA total of 18 female SD rats were randomize into sham group, model control group and the treatment group, with six rats in each group. Ovariectomy+STZ were used to establish the models. The treatment group was drenched with Qikui granules after modeling, sham group and model control group were intervened with the same amount of physiological saline. After three weeks of the intervention, to detect Glu, TG, BW and BMD of the rats in different groups, proliferation activity of MSCs was detected by MTT after culturing MSCs from rats in different groups in vitro, osteogenic differentiation ability of MSCs were determined by alizarin red staining; adipogenic differentiation ability of MSCs was detected by oil red O staining; the levels of B-ALP and BGP were measured by ELISA. ResultsAfter medicating for three weeks, compared with model control group, Glu lowered and BMD inereased in the treatment group (P<0.05); the number of mineralized calcium nodules was less in model control group (P<0.05); the number of adipocytes in model control group was more than that of the other two groups (P<0.05); after inducing the osteogenesis of BMSCs in different groups, the contents of BGP and B-ALP in model control group were lower than these of sham group notably (P<0.05), while Qikui granules could remarkably raise the levels of BGP and B-ALP in the model (P<0.05), and MTT suggested that the activity of cellular proliferation increased (P<0.05). ConclusionQikui granules could raise BMD in rat models when improving Glu, in-vitro cellular experiment indicates that Qikui granules could raise proliferation activity of MSCs in rat models, lift its osteogenic differentiation ability, at the same time, down regulate MSCs′ ability of differentiating into fat.
