摘要:
目的基于网络药理学方法研究芪参益气滴丸治疗缺血性脑卒中的作用机制。 方法从中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)数据库筛选芪参益气滴丸活性化合物及靶点,在蛋白质数据库Uniprot中转化为基因靶点,通过Cytoscape 3.7.1构建化合物-靶点网络。在线孟德尔人类遗传数据库(online mendelian inheritance in man,OMIM)及疾病相关的基因与突变位点数据库DisGeNET中收集缺血性脑卒中治疗靶点,构建疾病-靶点数据库。将化合物-靶点网络与疾病-靶点数据库结合,筛选芪参益气滴丸治疗缺血性脑卒中潜在核心靶点。利用蛋白质相互作用(PPI网络)数据库STRING,获得潜在核心靶点蛋白互作(PPI)网络。将潜在核心靶点进行基因本体论功能富集分析(gene ontology,GO)和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)/维基代谢通路(Wikipathway)富集分析。 结果芪参益气滴丸含有活性化合物68个,潜在核心靶点24个,包括NR3C2、KDR、PLAU、TP53、TNF、PTGS2、BIRC5、ITGB3、ESR1、PON1、PTGS1、IL4、ADRB2、TGFB1、F7、EDNRA、AR、ESR2、PPARG、NOS3、GRIA2、ADRA1A、LTA4H、MMP9,可能通过调节血管内皮生长因子信号通路、骨桥蛋白信号通路、内皮素信号通路、雌激素信号通路、炎性细胞因子介导的信号通路等影响活性氧代谢、一氧化氮合成、脂质代谢、炎症反应、神经血管重塑等机制,从而改善缺血性脑卒中病理损伤。 结论通过网络药理学可以预测芪参益气滴丸治疗缺血性脑卒中的潜在核心靶点、功能及相关信号通路,体现出多组分、多靶点、多机制的生物学效应。
Abstract:
ObjectiveTo study the mechanism of Qishen benefiting-Qi dripping pills in treating ischemic stroke based on network pharmacology. MethodsActive compound and targets of Qishen benefiting-Qi dripping pills were screened from TCMSP, and transformed into genetic targets in Uniprot database, to construct compound-target network via Cytoscape 3.7.1. Therapeutic targets of ischemic stroke were collected from OMIM and DisGeNET to build disease-targets database. The potential core targets of Qishen benefiting-Qi dripping pills in treating ischemic stroke were screened after combining compound-target network with disease-target database. STRING database was utilized to obtain the potential core target protein-protein interaction (PPI) network. The potential core targets were analyzed by GO, KEGG/Wikipathway. ResultsQishen benefiting-Qi dripping pills contained 68 active compounds and 24 potential core targets, including NR3C2, KDR, PLAU, TP53, TNF, PTGS2, BIRC5, ITGB3, ESR1, PON1, PTGS1, IL4, ADRB2, TGFB1, F7, EDNRA, AR, ESR2, PPARG, NOS3, GRIA2, ADRA1A, LTA4H and MMP9, the medicine could affect the metabolism of reactive oxygen species (ROS), nitric oxide synthesis, lipid metabolism, the inflammatory response, neurovascular remodeling possibly through adjusting vascular endothelial growth factor signaling pathway, osteopontin signaling pathway, endothelin signaling pathway, estrogen signaling pathway, inflammatory cytokines mediated signaling pathway, thereby improving pathological injury of ischemic stroke. ConclusionNetwork pharmacology could be used to predict the potential core targets, function and the relevant signaling pathway of Qishen benefiting-Qi dripping pills in the treatment of ischemic stroke, which embodies multi-component, multi-target and multi-mechanism biological effects.
