摘要:
目的运用网络药理学方法探究血府逐瘀汤治疗中晚期气滞血瘀型肺癌的作用机制。 方法通过中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)查询血府逐瘀汤的成分,并通过口服生物利用度(oral bioavailability,OB)≥30%,药物相似性(drug-likeness,DL)≥0.18,筛选化合物,通过查阅国内外文献和DrugBank数据库了解化合物的详细信息并进行确认核对。将核对后的化合物上传至Pharmmapper服务器并设置参数,通过Uniprot数据库、String数据库核对返回的靶点结果,最终得到血府逐瘀汤的有效靶点信息及蛋白互作网络图。通过在线孟德尔人类遗传数据库(online mendelian inheritance in man,OMIM)、GAD数据库、CTD数据库获得肺癌的相关靶点。利用韦恩图得出血府逐瘀汤和肺癌的共有靶点。将共有靶点导入Metascape数据库,设置物种为人类(sapiens),得到相关数据和PPI图。使用DAVID数据库、KOBAS数据库和Metascape数据库对共同靶点和信号通路进行基因本体论功能富集分析(gene ontology,GO)和KEGG富集分析(kyoto encyclopedia of genes and genomes,KEGG)并得出相应图标和结果。 结果从血府逐瘀汤中共筛选得到226个活性成分,其中重要成分有β-谷固醇、豆固醇、槲皮素、谷固醇、赤霉素、Shinpterocarpin等。这些成分对应262个靶点,其中与肺癌相关的人源靶点258个,关键靶点为TP53基因(tumor protein p53,TP53)、B淋巴细胞瘤2(B-cell lymphoma-2,Bcl-2)、表皮生长因子受体(epidermal growth factor receptor,EGFR)、丝氨酸/苏氨酸蛋白激酶(serine/threonine protein kinase,Akt1)、RELA及诱导型前列腺素内过氧化物合酶(prostaglandin-endoperoxide synthase 2,PTGS2)。通过DAVID数据库GO分析得出2833个主要生物学过程(biological process,BP),1521个细胞组成(cellular component,CC),2886个分子功能(molecular function,MF),KEGG、CORUM等多个通路富集库分析显示出1893条信号通路。其中肿瘤通路(hsa05200:Pathways in cancer)、白细胞介素4(interleukin-4,IL-4)和白细胞介素13(interleukin-13,IL-13)信号传导(R-HSA-6783783:Interleukin-4 and Interleukin-13 signaling)、信号传导(R-HSA-162582:Signal Transduction)、免疫系统(R-HSA-168256:Immune System)、白细胞介素的信号传递(R-HSA-449147:Signaling by Interleukins)等是较重要的通路。 结论血府逐瘀汤可能主要通过TP53、Bcl-2、EGFR、Akt1等靶点,通过hsa05200中的cAMP、丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)、PI3K-Akt、PPAR(peroxisome proliferater-activated receptor,PPAR)、血管内皮生长因子等多种信号通路抑制肿瘤生长、凋亡及转移来治疗中晚期气滞血瘀型肺癌。
Abstract:
ObjectiveTo discuss the mechanism of Xuefu Zhuyu Tang in the treatment of advanced lung cancer of Qi-stagnation blood-stasis pattern by using network pharmacology. MethodsThe components of Xuefu Zhuyu Tang were searched from TCMSP, and the compounds were screened by using oral bioavailability (OB)≥30%, drug likeness (DL)≥0.18, the detailed information of the compounds were surveyed by consulting the literature at home and abroad, and DrugBank database, confirmed and checked. The compounds after check were uploaded on to Pharmmapper server, to set the parameters, the targets returned were checked in Uniprot database and String database, consequently, to obtain the information of the effective targets of Xuefu Zhuyu Tang and PPI. The related targets of lung cancer were gained from OMIM database, GAD database and CTD database. Venn diagram was used to obtain the common targets of Xuefu Zhuyu Tang and lung cancer. After importing the targets into Metascape database, the species was set as sapiens, the relevant data and PPI diagram were obtained. DAVID database, KOBAS database and Metascape databases were used to perform GO enrichment and KEGG pathway enrichment analysis of the common targets and signaling pathway, therefore to obtain the corresponding icons and results. ResultsAll 226 active ingredients were screened and obtained from Xuefu Zhuyu Tang, among which, the important ingredients contained beta-sitosterol, stigmasterol, quercetin, sitosterol, Gibberellin A44 and Shinpterocarpin. The components corresponded to 262 targets, among them, 258 human-sourced targets were related to lung cancer, and the key targets were TP53, Bcl-2, EGFR, Akt1, RELA and PTGS2. GO analysis obtained 2833 BPs, 1521 CC, 2886 MF from DAVID database, the analysis of KEGG, CORUM and other pathways enrichment library displayed 1893 signaling pathways. Pathways in cancer, IL-4 and IL-13 signaling, signal transduction, immune system and signaling by interleukins are the crucial pathways. ConclusionXuefu Zhuyu Tang could inhibit the growth, apoptosis and metastasis of tumor possibly through the targets of TP53, Bcl-2, EGFR, Akt1 and other targets, cAMP, MAPK, P13K-Akt, PPAR, VEGF pathways in hsa05200, therefore to treat advanced lung cancer of Qi stagnation and blood stasis pattern.
