摘要:
目的基于临床处方信息，采用数据挖掘联合网络药理学方法剖析刘秀萍教授治疗慢性肾衰竭（chronic renal failure，CRF）的用药规律、核心药对及作用机制。 方法收集2018年12月至2021年12月间刘秀萍教授治疗CRF的临床处方并建库，采用数据挖掘方法分析处方中药使用频次、功效、四气五味及归经，并进行关联规则及聚类分析。利用网络药理学方法研究核心药对治疗CRF的作用机制。 结果共收录115个处方，包括128味中药，黄芪、白茅根、芦根、水蛭、三七、白术使用频数较高；四气五味以温、甘为首；归经以归肝、脾、肺经为主；药物功效以补虚为主，辅以活血化瘀、利水渗湿等；关联规则中“白茅根→黄芪”居支持度+置信度首位；聚类分析归为7类。核心药对作用于CRF的核心基因8个，主要通过作用于细胞外等细胞组分，参与癌症通路等通路，发挥酶结合等生物功能，参与一氧化氮生物合成过程的积极调节等生物过程。 结论宏观上得到刘秀萍教授治疗CRF的核心处方及药对；微观上发现核心药对活性成分，通过影响靶基因、信号通路等治疗CRF的作用机制，为刘教授治疗CRF中医理论提供了科学依据。
Abstract:
ObjectiveTo survey professor Liu Xiuping's medication laws, core drug pairs and the mecha-nism of treating CRF by using data mining and network pharmacology based on clinical prescriptions. MethodsProfessor Liu Xiuping's clinical prescriptions of treating CRF were collected between December, 2018 and December, 2021 to establish a database, data mining was used to analyze the frequency, efficacy, four natures and five properties and meridian tropism of the herbs in the prescription, to carry out association rules and cluster analysis. Network pharmacology was utilized to study the mechanism of the core drug pairs in the treatment of CRF. ResultsAll 115 prescriptions were included, containing 128 herbs, Huangqi (Astragali Radix), Baimaogen (Imperatae Rhizoma), Lugen (Phragmitis Rhizoma), Shuzhi (Hirudo), Sanqi (Notoginseng Radix ET Rhizoma) and Baizhu (Atractylodis Macrocephalae Rhizoma) were used in higher frequency, four natures and five flavors were of warm and sweet firstly; meridian tropism belonged to the ones of liver, spleen and lung; the effects of the drugs were tonification mainly, supplemented by promoting blood circulation and resolving stagnation, clearing damp and promoting diuresis; among the association rules, "Baimaogen-Huangqi" ranked the first in support+confidence; and its cluster analysis could be grouped into seven types. There were eight core genes of core drug acting on CRF, mainly through working on extracellular and other cellular components, participating in cancer pathways and developing biological functions such as enzyme binding, and participating in the active regulation of nitric oxide biosynthesis and other biological processes. ConclusionGenerally, professor Liu Xiuping's core prescriptions and drug pairs of treating CRF have been obtained; microscopically, the mechanism of active ingredients of core drug pairs in the treatment of CRF has been surveyed by influencing target genes and signaling pathways, which could provide scientific reference for professor Liu Xiuping's TCM theory of treating CRF.
