目的研究益气养阴活血法对特发性肺纤维化(idiopathic pulmonary fibrosis,IPF)大鼠血清低氧诱导因子1α(hypoxia inducible factor-1α,HIF-1α)、血小板源性生长因子(platelet-derived growth factor,PDGF)、色素上皮衍生因子(pigment epithelium derived factor,PEDF)和内皮生长抑制素(endostatin,ES)的影响。 方法将128只大鼠随机分为空白组、模型组、泼尼松组、参七虫草组,每组32只,采用气管滴注博来霉素方法进行IPF造模,第7、14、21、28天检测大鼠血清HIF-1α、PDGF、PEDF、ES含量。 结果造模后第7、14、21、28天,模型组、泼尼松组、参七虫草组大鼠血清HIF-1α、PDGF、PEDF、ES含量高于空白组(P<0.01);与模型组比较,泼尼松组与参七虫草组各时间点大鼠血清HIF-1α、PDGF含量降低(P<0.01),PEDF、ES表达升高(P<0.01);各时间点大鼠血清各指标泼尼松组与参七虫草组比较差异无统计学意义(P>0.05)。 结论益气养阴活血法可干预IPF大鼠肺纤维化进程,机制可能与调节血管新生相关因子从而抑制血管新生有关。
Abstract:
ObjectiveTo discuss the influence of benefiting-Qi nourishing-Yin activating-blood method on HIF-1α, PDGF, PEDF and ES in rats suffering idiopathic pulmonary fibrosis (IPF). MethodsAll 128 rats were randomized into the blank group, the model group, prednison group and Shenqi Chongcao group, with 32 ones in each group, tracheal dripping of bleomycin was adopted to carry out IPF modeling, to detect the contents of HIF-1α, PDGF, PEDF and ES in rat serum on the 7th, 14th, 21st and 28th day. ResultsOn the 7th, 14th, 21st and 28th day, the contents of HIF-1α, PDGF, PEDF and ES in the model group, prednison group and Shenqi Chongcao group increased, but higher than these of the blank group (P<0.01); compared with the model group, the contents of HIF-1α and PDGF in prednison group and Shenqi Chongcao group were lower at different time points (P<0.01). The contents of PEDF and ES of the model group, prednison group and Shenqi Chongcao group were higher than these of the blank group (P<0.01); The different showed no statistical meaning in different indeses between prednison group and Shenqi Chongcao group at different time points (P>0.05). ConclusionBenefiting-Qi nourishing-Yin activating-blood method could intervene the process of pulmonary fibrosis of IPF rats, and its mechanism might be related to regulating angiogenesis associated factors, thus inhibiting angiogenesis.
