目的采用网络药理学方法分析补骨脂-淫羊藿药对抗绝经后骨质疏松症(postmenopausal osteoporosis,PMOP)的作用机制。 方法利用TCMSP数据库,TCM Database@Taiwan数据库和BATAMAN-TCM数据库检索并筛选补骨脂和淫羊藿的有效成分,然后预测有效成分的靶蛋白,构建有效成分-靶蛋白网络。利用GeneCards数据库,OMIM数据库,GKB数据库和TTD数据库检索PMOP的相关蛋白和基因,找到补骨脂-淫羊藿药对治疗PMOP的相关靶点,进行富集分析。 结果补骨脂-淫羊藿药对共获得51个有效成分和292个靶蛋白,其中节点度>5的共有21个有效成分与26个靶蛋白相连,其中槲皮素,山柰酚,木犀草素,异补骨脂甲素和脱水淫羊藿素处于核心位置。对补骨脂-淫羊藿药对与PMOP共有的132个共同靶标分析显示,共同靶点主要富集在转录因子活性、类固醇激素受体活性、蛋白激酶结合和营养水平等,主要富集的信号通路有糖尿病并发症中的AGE-RAGE信号通路、癌症相关途径、流体剪切应力与动脉粥样硬化、内分泌耐药和PI3K-Akt信号通路。 结论补骨脂-淫羊藿药对可能通过AGE-RAGE信号通路、流体剪切应力、PI3K-Akt信号通路和调节内分泌系统多靶点、多途径治疗PMOP。
Abstract:
ObjectiveTo analyze the mechanism of Buguzhi(Psoraleae fructus)-Yinyanghuo (Epimedii folium) against postmenopausal osteoporosis (PMOP) using network pharmacology. MethodsTCMSP database, TCM database@Taiwan database and BATAMAN-TCM database were utilized to search and screen the active ingre-dients of Buguzhi and Yinyanghuo, and then predict the target protein of the active ingredients, construct the active ingredients-target protein network. GeneCards database, OMIM database, GKB database and TTD database were applied to retrieve the related protein and genes of PMOP, find out the relevant targets of the couplet medicines in the treatment of PMOP, and perform the enrichment analysis. ResultsAll 51 active ingredients and 292 target proteins were gained from the drug combination, among them, 21 active components with degree>5 were linked to 26 target proteins, quercetin, kaempferol, luteolin, isobavachin and anhydroicaritin are at the core. The analysis of 132 common targets shared by Buguzhi-Yinyanghuo with PMOP showed that the common targets were mainly enriched in the transcription factor activity, steroid hormone receptor activity, protein kinase binding, and nutritional levels, etc, and the main enriched signaling pathways contained AGE-RAGE signaling pathway in diabetic complications, cancer related pathway, fluid shear stress and atherosclerosis, endocrine resistance and PI3K-Akt signaling pathway. ConclusionBuguzhi-Yinyanghuo may treat PMOP with multi-targets and multi-pathways, possibly through the AGE-RAGE signal pathway, fluid shear stress, PI3K-Akt signal pathway and regulating endocrine system.
