摘要:
目的探究益气养阴方联合电针对产后压力性尿失禁(postpartum urinary incontinence,PPUI)大鼠趋化因子受体4(chemokine receptor 4,CXCR4)/环氧合酶2(cyclooxygenase,COX-2)信号通路的作用机制。 方法将大鼠随机分为空白组、PPUI组、益气养阴组和益气养阴+电针组,制备PPUI大鼠模型;检测4组大鼠收缩力/肌重比值、血清乳酸脱氢酶(lactate dehydrogenase,LDH)和心肌酶肌酸激酶(creatine kinase,CK)含量;HE染色检测4组大鼠肛提肌组织病理学变化;蛋白质印迹法检测4组大鼠尿道组织CXCR4和COX-2蛋白表达。 结果与空白组相比,大鼠最大膀胱容量和漏尿压力点PPUI组降低(P<0.05),益气养阴组和益气养阴+电针组升高,益气养阴+电针组升高明显(P<0.05);与空白组相比,收缩力/肌重比值PPUI组降低(P<0.05),益气养阴组和益气养阴+电针组较PPUI组增加,且益气养阴+电针组增加更明显(P<0.05);与空白组相比,LDH和CK含量PPUI组升高(P<0.05),益气养阴组和益气养阴+电针组低于PPUI组(P<0.05)。与空白组相比,PPUI组CXCR4蛋白减少,COX-2蛋白增多(P<0.05),益气养阴组和益气养阴+电针组CXCR4蛋白较PPUI组增多,COX-2蛋白较PPUI组减少(P<0.05),益气养阴+电针组CXCR4蛋白较益气养阴组增加,COX-2蛋白较益气养阴组减少(P<0.05)。 结论益气养阴方联合电针可能通过抑制PPUI大鼠血清LDH和CK表达,增加PPUI大鼠CXCR4表达,抑制COX-2表达,增加盆底肌肌肉收缩功能,治疗PPUI。
Abstract:
ObjectiveTo survey the mechanism of benefiting-Qi nourishing-Yin prescription in combination with electroacupuncture on CXCR4/COX-2 signal pathway in rats suffering postpartum urinary incontinence (PPUI). MethodsAll the rats were randomized into the blank group, PPUI group, benefiting-Qi nourishing-Yin group, benefiting-Qi nourishing-Yin+electroacupuncture group, to establish PPUI rat models; to detect contraction force/muscle weight ratio of the rats in four groups. The contents of LDH and CK; HE staining is applied to detect histopathological changes of musculi levator ani of the rats in four groups; Western blotting method to measure the expressions of CXCR4 and COX-2 protein in urethral tissue of the rats in four groups. ResultsCompared with the blank group, maximum bladder capacity and leakage pressure point of rats in PPUI group were decreased (P<0.05), after intervention, maximum bladder capacity and leakage pressure point of benefiting-Qi nourishing-Yin group and benefiting-Qi-Yin+electroacupuncture group were significantly restored, and the recovery in benefiting-Qi nourishing-Yin+electroacupuncture group was the best (P<0.05); compared with the blank group, the ratio of contractile force to muscle weight in the PPUI group was lowered (P<0.05), the ratio in benefiting-Qi nourishing-Yin group and benefiting-Qi nourishing-Yin+electroacupuncture group was significantly higher than that in the PPUI group, and the increase in benefiting-Qi nourishing-Yin+electroacupuncture group was more noticeable (P<0.05); compared with the blank group, the contents of LDH and CK increased in the PPUI group (P<0.05), benefiting-Qi nourishing-Yin group and benefiting-Qi nourishing-Yin+electroacupuncture group were lower than the PPUI group (P<0.05). Compared with the blank group, CXCR4 protein reduced while COX-2 protein increased in the PPUI group (P<0.05), the increase of CXCR4 protein in benefiting-Qi nourishing-Yin group and benefiting-Qi nourishing-Yin+electroacupuncture group were more than that in the PPUI group, COX-2 protein reduction was more obvious than that in the PPUI group (P<0.05), the increase of CXCR4 protein in benefiting-Qi nourishing-Yin+electroacupuncture group was higher than that in benefiting-Qi nourishing-Yin group, the contents of COX-2 protein were less than those in benefiting-Qi nourishing-Yin group (P<0.05). ConclusionBenefiting-Qi nourishing-Yin prescription in combination with electroacupuncture could increase the expressions of CXCR4 while restrain the expressions of COX-2 in PPUI rats, the contraction of pelvic floor muscle and treat PPUI possibly through inbibiting the expressions of LDH and CK in PPUI rats.
