摘要:
目的探讨马里苷对糖尿病小鼠肝脏、胰腺的组织形态以及自噬调节的影响。 方法选择10只db/m小鼠为对照组，按照随机对照原则将另外30只db/db小鼠分为模型组、二甲双胍阳性药组和马里苷给药组，每组10只。各组连续灌胃给药8周，HE染色观察各组小鼠肝脏和胰腺的形态学变化、免疫组化检测小鼠肝脏P62的含量、Western Blotting检测小鼠肝脏自噬相关蛋白LC3Ⅱ/I、P62、ATG5和Beclin1的表达水平。 结果组织形态学显示，二甲双胍和马里苷给药组小鼠肝小叶结构较清晰，肝索排列较紊乱，肝细胞气球样变有所减轻，且胰岛形态较正常，边界比较清晰，胰岛细胞排列有明显改善，胰岛细胞空泡状变性减少。肝脏免疫组化结果显示，模型组小鼠P62表达高于对照组，二甲双胍阳性药组和马里苷给药组使病理状态下P62的表达明显降低。蛋白免疫印迹结果显示，与模型组比较，马里苷和二甲双胍可以显著增加糖尿病小鼠肝脏自噬相关蛋白LC3Ⅱ/I、ATG5和Beclin1的表达（P＜0.05），降低P62的表达（P＜0.05）。 结论马里苷可缓解糖尿病所造成的肝脏和胰腺的组织病理损伤；促进糖尿病小鼠肝脏自噬表达增强。
Abstract:
ObjectiveTo discuss the influence of marein on the histomorphology of liver and pancreas, autophagy regulation in diabetic mice. MethodsTen db/m mice were chosen as the control group, another 30 db/m mice were selected as the model group, metformin-positive medicine group and marein-administered group according to random controlled principles with ten in each group. All the groups accepted intragastric administra-tion for eight weeks continuously, HE staining was used to observe the morphology of liver and pancreas of the mice in different groups, immunohistochemical method to detect the contents of P62 in the liver of the mice, Western Blotting method to measure the expressions of autophagy-related proteins LC3Ⅱ/I, P62, ATG5 and Beclin1 in the liver of the mice. ResultsThe histomorphology showed that the mice of metformin-positive medicine group and marein-administered group had a clearer structure of the liver lobules, with more disorganized hepatic cords, relieved hepatocyte ballooning, normal islet morphology, having clearer borders, there was a significant improvement in the arrangement of islet cells, and a reduction in islet cell vacuolar degeneration. Liver immunohistochemistry revealed that the expressions of P62 in the model group were higher than these of the control group, the expressions of P62 were reduced clearly under the pathological state in metformin-positive medicine group and marein-administered group. The results of Western Blotting indicated that, compared with the model group, marein and metformin could noticeably increase the expressions of autophagy-related proteins LC3Ⅱ/I, ATG5 and Beclin1 in the liver of the mice (P<0.05), decrease the expressions of P62 (P<0.05). ConclusionMarein could reduce the histopathological damage of liver and pancreas caused by the diabetes; and enhance the expressions of liver autophagy genes in diabetic mice.
