摘要:
目的运用网络药理学探索益肾健脾中药“山药-杜仲”药对治疗骨质疏松症的作用机制。 方法通过中药数据库检索“山药-杜仲”的活性成分及相关作用靶点,运用Cytoscape 3.7.2制作“活性成分-靶点”网络图。利用GeneCards数据库搜集骨质疏松症相关靶点,将药物靶点与疾病靶点进行映射,取交集靶点上传STRING 11.0数据库构建蛋白互作网络,将该网络导入Cytoscape 3.7.2进一步加工分析。将关键靶点上传至Metascape平台进行GO富集分析与KEGG通路富集分析。 结果共获得山药和杜仲活性成分38个,对应靶点225个,骨质疏松症相关靶点1376个。药物-疾病作用核心靶点80个,包括白蛋白、白细胞介素6、腺苷酸激酶同工酶1、血管内皮生长因子、抑癌基因p53、肿瘤坏死因子、基质金属蛋白酶9等。KEGG富集分析显示其作用于骨质疏松症的潜在通路主要包括糖尿病并发症的AGE-RAGE信号通路、T细胞受体信号通路、p53信号通路、TGF-β信号通路、HIF-1信号通路等。 结论本研究初步探讨了益肾健脾药对“山药-杜仲”对骨质疏松症的“多通路、多靶点”作用机制,为进一步探索益肾健脾法治疗骨质疏松症提供研究思路与理论基础。
Abstract:
ObjectiveTo explore the mechanism of benefiting-kidney invigorating-spleen herbs Chinese yam-Duzhong (eucommiae cortex) in the treatment of osteoporosis using network pharmacology. MethodsThe active ingredients and related targets of Chinese yam-Duzhong were searched by TCM database (TCMSP), and the network diagram of "active ingredients-targets" was made by using Cytoscape3.7.2. The targets related to osteoporosis were collected using GeneCards database, the intersection targets were obtained by mapping the drug targets to the disease targets, and uploaded to STRING 11.0 database to build PPI, which was imported into Cytoscape 3.7.2, processed and analyzed. The key targets were uploaded to Metascape platform to carry out GO enrichment analysis and KEGG pathway enrichment analysis. ResultsThe study obtained 38 active ingredients from Chinese yam and Duzhong, 225 corresponding targets and 1376 targets related to the disease. There were 80 core targets of medicine-disease action, including albumin, interleukin 6 (IL6), adenylate kinase isoenzyme 1, vascular endothelial growth factor (VEGF), tumor suppressor p53, tumour necrosis factor, MMP9 and others. KEGG enrichment analysis displayed that the potential pathway acting on osteoporosis mainly included AGE-RAGE signal pathways of diabetic complication, T cell receptor signal pathway, p53 signal pathway, TGF-β signal pathway and HIF-1 signal pathway. ConclusionThis study has primarily discussed the "multi-pathway, multi-target" mechanism of Chinese yam-Duzhong, the couplet medicines which could benefit the kidney and invigorate spleen, in the treatment of osteoporosis, wishing to provide the research thinking and theoretical foundation for further exploration into the treatment of osteoporosis by benefiting-kidney invigorating-spleen method.
