目的基于网络药理学及分子对接分析参苓白术散治疗特应性皮炎(atopic dermatitis,AD)的作用机制。 方法运用系统药理学方法查找并筛选参苓白术散方中各中药的活性成分及靶点,查找AD相关基因,收集参苓白术散治疗AD的靶点,进行富集分析以预测其可能的信号通路,并以分子对接技术进行验证。 结果筛选得到参苓白术散活性成分153个及AD相关的靶点1600个,根据交集靶点构建蛋白-蛋白互作网络图并筛选得到9个核心靶点,基因本体富集分析发现其功能主要与维生素D生物合成过程的正调控、降钙素1-单加氧酶活性的正调控、小窝、基底质膜、树突状细胞质、质膜筏等有关,京都基因与基因组百科全书主要富集在癌症的通路等,分子对接显示活性成分槲皮素、木犀草素与JUN、MAPK结合紧密。 结论参苓白术散治疗AD具有多途径、多靶点、多层次的特点,其作用机制复杂,为后续实验验证提供理论依据。
Abstract:
ObjectiveTo analyze the mechanism of Shenling Baizhu powder in the treatment of atopic dermatitis (AD) based on network pharmacology and molecular docking. MethodsThe method of systematic pharmacology was used to search and screen the active ingredients and the targets of the herbs contained in the powder, to search for AD-related genes, and to collect the targets of the powder in the treatment of AD for the enrichment analysis to predict the potential signaling pathways, and molecular docking was applied for validation. ResultsAll 153 active ingredients of Shenling Baizhu powder and 1600 AD-related targets were obtained, PPI was built according to the intersected targets and nine core targets were screened and gained, GO enrichment analysis revealed that its functions are primarily associated with the positive regulation of vitamin D biosynthesis, the positive regulation of calcitonin 1-monooxygenase activity, the small vacuole, the basal membrane, the cytoplasm of dendritic cells and the plasma membrane raft. KEGG is primarily enriched in cancer pathways, and molecular docking showed that active compounds such as quercetin and luteolin are closely combined with JUN and MAPK. ConclusionShenling Baizhu powder in the treatment of AD has the characteristics of multiple pathways, multiple targets and multiple levels with the complicated mechanism of action, which could provide theoretical basis for subsequent experimental verification.
