摘要:
目的基于血管生成素1(angiopoietin-1,Ang-1)/酪氨酸激酶受体2(tyrosine kinase receptor-2,Tie-2)信号通路探讨生地黄梓醇对缺血性脑卒中大鼠的促血管新生与神经功能重塑作用。 方法将24只清洁级雄性SD大鼠随机分为假手术组、模型组、梓醇干预组,每组8只。梓醇干预组和模型组采取Longa改良线栓法造模,假手术组仅分离颈动脉,不插入线栓。梓醇干预组于造模24 h后腹腔注射9 mg/(kg·d)生地黄梓醇溶液进行干预,假手术组和模型组大鼠常规饲养并腹腔注射等量的生理盐水,每日1次,连续干预7天。分别在造模后第1、5、8天,对各组大鼠进行神经功能评分和粘签试验。在造模后第8天,用TTC染色法观察大鼠脑组织形态学变化,qRT-PCR法检测Ang-1、Ang-2、Tie-2 mRNA表达情况。 结果时间和组别在双侧粘签去除时间结果方面存在交互作用(P<0.05);时间和组别在双侧粘签去除时间结果方面主效应显著(P<0.05)。在造模后第1天,与假手术组相比,模型组和梓醇干预组大鼠神经功能评分、双侧贴纸去除时间高于假手术组(P<0.05);与模型组相比,造模后第5、8天梓醇干预组大鼠神经功能评分、双侧贴纸去除时间降低(P<0.05)。与造模后第1天相比,梓醇干预组大鼠在造模后第5、8天神经功能评分、双侧贴纸去除时间降低(P<0.05)。与造模后第5天相比,梓醇干预组大鼠在造模后第8天神经功能评分、双侧贴纸去除时间降低(P<0.05)。TTC染色显示假手术组大鼠没有出现脑梗死区域,模型组和梓醇干预组大鼠均有明显脑梗死区域,但梓醇干预组干预后脑梗死范围较模型组缩小。与模型组相比,梓醇干预组大鼠Ang-1、Tie-2 mRNA表达水平升高(P<0.05);模型组与梓醇干预组大鼠Ang-2 mRNA表达水平比较,差异无统计学意义(P>0.05)。 结论生地黄梓醇可通过上调Ang-1、Tie-2 mRNA水平促血管新生与神经功能重塑,两者协同可发挥促血管新生而不增加局部组织水肿的作用。
Abstract:
ObjectiveTo discuss the effects of promoting angionesis and neural remodeling of catalpol on ischemic stroke rats based on Ang-1/Tie-2 signaling pathway. MethodsA total of 24 male SD rats were randomized into sham operation group, the model group and catalpol intervention group with eight in each group. Catalpol intervention group and the model group were established using the modified suture occlusion method, for the sham operation group, only carotid artery separation was performed, with no guidewire inserted. Catalpol intervention group received catalpol via intraperitoneal injection at a dose of 9 mg/(kg·d) 24 h after modelling, the sham operation group and the model group received conventional feeding and intraperitoneal injection of the same amount of saline, once each day, for seven consecutive days. Neurological function scoring and adhesion testing were conducted in each group on the first, fifth and eighth day after modeling. TTC staing method was applied to observe cerebral tissue morphological changes of the rats on the eighth day after modeling, qRT-PCR was used to detect the exprssions of Ang-1, Ang-2 and Tie-2 mRNA. ResultsSignificant time × group interaction was observed for bilateral adhesive label removal time (P<0.05); time and group both had a significant main effect on the removal time of bilateral adhesive labels (P<0.05). On the first day after modeling, compared with sham operation group, the model group and catalpol intervention group were higher in neurological function scores and the removal time of bilateral adhesive labels (P<0.05); compared with the model group, neurological function scores and the removal time of bilateral adhesive labels were reduced in catalpol intervention group on the fifth and eighth day after modeling (P<0.05). Compared with the first day after modeling, neurological function scores and the removal time of bilateral adhesive labels were lower in catalpol intervention group on the fifth and eighth day after modeling (P<0.05). Compared with the fifth day after modeling, neurological function scores and the removal time of bilateral adhesive labels were lower in catalpol intervention group on the eighth day after modeling (P<0.05). TTC staing method revealed that no cerebral infarction was found in the sham operation group, obvious cerebral infarction area was observed in the model group and catalpol intervention group, however, the area of cerebral infarction diminished after catalpol intervention compared with the model group. The levels of Ang-1 and Tie-2 mRNA were elevated in catalpol intervention group than the model group (P<0.05); The difference had no statistical meaning in the expressions of Ang-2 mRNA between the model group and catalpol intervention group (P>0.05). ConclusionCatalpol could promote angionesis and neural remodeling via upregulating the levels of Ang-1 and Tie-2 mRNA, the synergy between the two could promote angionesis without increasing local tissue edema.
