目的探讨槐花金银酒对黑色素瘤小鼠免疫调节作用及其机制。
方法选取14只C57BL/6小鼠构建荷瘤模型,造模后分为溶剂对照组与槐花金银酒组,每组7只。溶剂对照组灌胃给予0.5%羧甲基纤维素钠,槐花金银酒组灌胃给予槐花金银酒提取物,两组均干预14天。监测两组小鼠的肿瘤体积、肿瘤质量及体质量;采用流式细胞术检测两组荷瘤小鼠胸腺和淋巴结中免疫细胞[辅助性T细胞(T helper cells,Th)、杀伤T细胞(cytotoxic T cells,Tc)、树突状细胞(dendritic cells,DC)、自然杀伤细胞(natural killer cells,NK)及调节性T细胞(regulatory T cells,Treg)]的占比;采用酶联免疫吸附法检测两组荷瘤小鼠胸腺和淋巴结组织悬液中免疫细胞因子[白细胞介素6(interleukin-6,IL-6)、IL-10及转化生长因子β(transforming growth factor-β,TGF-β)]的水平。
结果与溶剂对照组相比,槐花金银酒组小鼠肿瘤体积、肿瘤质量均减小(P<0.05),小鼠体质量无明显变化(P>0.05),胸腺和淋巴结中Th细胞、Tc细胞、DC细胞和NK细胞占比升高(P<0.05),Treg细胞占比及IL-6、IL-10、TGF-β水平均降低(P<0.05)。
结论槐花金银酒可通过调节机体免疫功能抑制黑色素瘤生长,且对小鼠体质量无明显影响,具有一定安全性。

ObjectiveTo explore the immunoregulation activities of sophora flower (Sophorae flos)-honeysuckle (Lonicerae japonicae flos) liquor on melanoma mice and its mechanism.
MethodsA total of 14 C57BL/6 mice were chosen to construct the tumor-bearing mice models, and divided into solvent control group and sophora flower-honeysuckle liquor group with seven in each after successfully modeling. Solvent control group were drenched with 0.5% sodium carboxymethylcellulose, and the extract of sophora flower-honeysuckle liquor was given to sophora flower-honeysuckle liquor group by intragastric administration, and both groups were intervened for 14 days. To monitor tumor volume, tumor weight and body mass of the mice in two groups; flow cytometry was adopted to detect the proportions of immunological cells (Th, Tc, DC, NK and Treg) in thymus gland and lymph node of tumor-bearing mice in the two groups; ELISA was applied to measure the levels of immune cytokines such as IL-6, IL-10 and TGF-β in the suspension of thymus gland and lymph node of the tumor-bearing mice in the two groups.
ResultsCompared with solvent control group, tumor volume and tumor weight reduced in sophora flower-honeysuckle liquor group (P<0.05), no significant changes in body mass of the mice were observed in the mice (P>0.05), the proportions of Th, Tc, DC and NK cells increased in thymus gland and lymph nodes (P<0.05), the proportion of Treg cells and the levels of IL-6, IL-10 and TGF-β reduced (P<0.05).
ConclusionSophora flower-honeysuckle liquor group could inhibit the growth of melanoma by regulating the body’s immune function, showing no significant effects on the body mass of mice and the safety to a certain extent.
