摘要:
目的运用网络药理学及分子对接方法探讨归脾汤基于肠道菌群治疗血管性痴呆(vascular dementia,VD)的可能作用机制。 方法从BATMAN-TCM/ETCM数据库筛选归脾汤的活性成分,利用Pubchem、SwissADME及Swiss Target Prediction平台筛选出有效活性成分及对应靶点,利用UniProt数据库规范靶点;在人类基因数据库(the human gene database,GeneCards)、在线人类孟德尔遗传数据库(online mendelian inheritance in man,OMIM)筛选VD及肠道菌群主要靶点,取归脾汤-VD疾病交集靶点并在Cytoscape软件中构建和分析归脾汤-活性成分-靶点网络;运用STRING平台构建蛋白-蛋白互作(protein-protein interactions,PPI)网络,cytoHubba插件筛选Hub基因;基于核心靶基因导入基因功能注释数据库(the database for annotation visualization and integrated discovery,DAVID)平台进行基因本体论(gene ontology,GO)功能和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)通路富集分析,最后用Autodock进行分子对接验证。 结果归脾汤有鼠李糖苷、3,3’-二甲基槲皮素、熊果菊素、山柰酚等205个有效活性成分;TNF、MMP9、IL-6、PTGS2、JUN等35个靶点与VD、肠道菌群相关;GO功能富集主要涉及对氧化应激、炎症反应、细胞增殖的调控等相关生物进程,富集于脂质和动脉粥样硬化、IL-17、AGE-RAGE信号通路在糖尿病并发症的应用等相关通路;分子对接活性均较好,其中MMP9与Rhamnocitrin对接最佳。 结论归脾汤通过多靶点、多通路抑制血管内皮活化,参与肠道微生物重组与肠道屏障重建,并通过缓解肠道炎症直接或间接影响中枢神经系统。此外,肠道菌群对脂质及糖的代谢也为其治疗发挥辅助作用。
Abstract:
ObjectiveTo explore the possible mechanism of Guipi decoction in treating vascular dementia (VD) on the foundation of gut microbiota using network pharmacology and molecular docking. MethodsActive ingredients of the decoction were screened from BATMAN-TCM/ETCM, Pubchem, SwissADME and Swiss Target Prediction platform were utilized to screen active ingredients and the corresponding targets, and UniProt was employed to standardize the targets; the main targets of VD and gut microbiota were searched from GeneCards and OMIM, to take the intersecting targets between Guipi decoction-VD, to construct and analyze Guipi decoction-active ingredient-target network using Cytoscape software; STRING platform was applied to build PPI network, cytoHubba plug-in to identify Hub gene; based on the core target genes, GO and KEGG pathway enrichment analysis were performed using DAVID, consequently, molecular docking validation was conducted with Autodock. ResultsAll 205 active ingredient including rhamnoside, 3, 3’-dimethylquercetin, eriodictyol and kaempferol were identified in the decoction; 35 targets covering TNF, MMP9, IL-6, PTGS2 and JUN were related to VD and gut microbiota; GO functional enrichment mainly involved BP such as oxidative stress, inflammatory reaction and the regulation of cellular proliferation, enriched in the application of lipid and atherosclerosis, IL-17 and AGE-RAGE signaling pathway in diabetic complications; molecular docking results demonstrated promising binding affinity, among them, MMP9 exhibited the best binding with Rhamnocitrin. Conclusion Guipi decoction could inhibit vascular endothelial activation via multi-target and multi-pathway, participate in the remodeling of the gut microbiota and restoration of the intestinal barrier, directly or indirectly affect central nervous system by relieving intestinal inflammation. Additionally, gut microbiota - mediated lipid and glucose metabolism exerts therapeutic effects on VD.
