摘要:
目的研究白术内酯Ⅰ对白塞病小鼠脾脏辅助性T细胞17(T helper cells 17,Th17)/调节性T细胞(regulatory T cell,Treg)平衡的影响。 方法将60只BALB/c小鼠随机分为空白组、模型组和实验组,每组20只,参考文献方法进行造模。空白组与模型组小鼠给予等体积0.9%的生理盐水,实验组给予白术内酯Ⅰ干预。采用流式细胞术检测小鼠脾脏中Th17和Treg细胞数量;采用酶联免疫吸附(enzyme-linked immunosorbent assay,ELISA)法检测各组小鼠脾脏中Th17细胞因子白细胞介素6(interleukin-6,IL-6)、IL-17水平及Treg细胞因子IL-4和IL-10的表达水平;采用蛋白免疫印迹法检测小鼠脾脏中Th17关键调控因子维甲酸相关孤儿受体γt(retinoic acid-related orphan receptor gamma t,RORγt)及Treg关键调控因子叉头/翼状螺旋转录因子3(forkhead box p3,Foxp3)的表达水平。 结果与空白组相比,模型组小鼠Th17、IL-6、IL-17、RORγt表达水平及Th17/Treg比值均升高(P<0.05);IL-10、Treg、Foxp3表达水平均降低(P<0.05)。与模型组相比,实验组小鼠Th17、IL-6、IL-17、RORγt表达水平及Th17/Treg比值均降低(P<0.05);IL-10、Treg及Foxp3表达水平均升高(P<0.05)。 结论白术内酯Ⅰ可能通过调节Th17/Treg免疫失衡,改善原肌球蛋白诱导的白塞病小鼠的免疫功能。
Abstract:
ObjectiveTo study the effects of atractylenolide I on spleen Th17/Treg balance in Beh?ets disease (BD) mice. MethodsSixty mice were randomly assigned to three groups: a blank control group, a model group and an experiment group with 20 in each. The models were then established following the method described in the literature. Mice in the blank control group and the model group were administered an equal volume of 0.9% saline, while the experiment group received atractylenolide I. Flow cytometry was adopted to detect the number of Th17 and Treg cells in the spleen; ELISA method was employed to measure the levels of Th17 cytokines IL-6, IL-17, and the expressions of Treg cytokines IL-4 and IL-10 in the spleen of the mice in all the groups; Western blotting method was applied to determine the levels of critical Th17 regulator RORγt and critical Treg regulator Foxp3 in the spleen of the mice. ResultsCompared with the blank control group, the increased expressions of Th17, IL-6, IL-17 and RORγt, and Th17/Treg ratio was observed (P<0.05); while the downregulation of IL-10, Treg and Foxp3 was detected in the model group (P<0.05). Compared with the model group, the experiment group exhibited the decreased expressions of Th17, IL-6, IL-17 and RORγt, and Th17/Treg ratio (P<0.05); and the upregulation of IL-10, Treg and Foxp3 (P<0.05). ConclusionAtractylenolide I could improve immune function in tropomyosin-induced BD mice possibly through regulating Th17/Treg imbalance.
