摘要:
目的探讨水蛭治疗心肌梗死(myocardial infarction,MI)的差异靶基因和信号通路。 方法从基因表达综合数据库(gene expression omnibus,GEO)中检索与MI有关的数据芯片,利用GEO2R工具对数据芯片进行在线分析,筛选出MI差异表达的靶基因;利用中医药综合数据库(traditional Chinese medicine integrated database,TCMID)、比较毒理基因组学数据库(comparative toxicogenomics database,CTD)、BATMAN、SwissTargetPrediction数据库筛选与水蛭相关的靶点;利用STRING数据库和核心靶基因导入基因功能注释数据库(the database for annotation visualization and integrated discovery,DAVID),对水蛭治疗MI的差异靶基因进行基因本体论(gene ontology,GO)富集分析和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)富集分析。 结果利用GEO2R工具筛选出794个与MI有关的差异靶基因,包括386个下调基因,408个上调基因;利用TCMID数据库得到518个与水蛭相关的靶点;最终筛选出28个水蛭治疗MI的差异靶基因,10个关键基因;经GO和KEGG富集分析得到P<0.05的生物过程(biological process,BP)25个、细胞组分(cell component,CC)5个、分子功能(molecular function,MF)27个,信号通路22条。 结论水蛭治疗MI的差异靶基因包括VEGFA、AREG、EGFR等基因,其作用机制可能与水蛭调控Rap1、PI3K-Akt、Ras、MAPK、HIF-1等信号通路有关。
Abstract:
ObjectiveTo explore the differential target genes and signaling pathways of leech in the treatment of myocardial infarction (MI). MethodsMI-related data chips were searched from GEO database, GEO2R tools were applied to carry out online analysis of data chips, and to screen the differential target genes of MI; TCMID, CTD, BATMAN, SwissTargetPrediction data were utilized to screen leech-related targets; GO enrichment analysis and KEGG enrichment analysis of the differential target genes of leech in the treatment of MI were performed using the STRING database and core target genes imported into DAVID. ResultsGEO2R tools were used to screen 794 differential target genes related to MI, including 386 down-regulated genes and 408 up-regulated genes; TCMID was applied to obtain 518 leech-related targets; 28 differential target genes and ten core genes were selected; 25 biological process with P<0.05, five CC, 27 MF and 22 signaling pathways were yielded by GO and KEGG enrichment analysis. ConclusionThe differential genes of leech for MI include VEGFA, AREG, EGFR, etc, and its mechanism might be related to the regulation of the signaling pathways including Rap1, PI3K-Akt, Ras, MAPK and HIF-1 by leech.
