摘要:
目的：研究骨痹止痛液对动物模型膝关节软骨中白细胞介素-1(IL-1)、肿瘤坏死因子α(TNF-α)的影响。方法：将30只新西兰大白兔随机分为骨痹止痛液组(A组)、盐酸氨基葡萄糖片组(B组)、模型组(C组)3组，每组10只。建立膝骨性关节炎模型后，分别给予骨痹止痛液、盐酸氨基葡萄糖片、生理盐水灌胃，连续8周。于8周后取样，采用肉眼观察形态学改变、HE染色、RT-PCR检测软骨中IL-1、TNF-α因子表达等指标。结果：肉眼观察：A组及B组较给药前无明显变化，C组关节软骨破坏较为严重，甚至出现糜烂。RT-PCR：A组、B组与C组比较，A组、B组兔软骨标本中IL-1mRNA、TNF-α mRNA表达量明显降低(P＜0.05)。兔软骨标本中IL-1mRNA、TNF-α mRNA表达量A组较B组减低(P＜0.05)。结论：骨痹止痛液通过减少促炎因子(IL-1、TNF-α)的表达来抑制炎症反应，从而减缓关节损伤。
Abstract:
Objective: To study the influence of GuBi ZhiTong liquid on IL-1 and TNF-α in knee cartilage of the animal model. Methods: Thirty rabbits were randomized into the group of GuBi ZhiTong liquid (A), the group of glucosamine hydrochloride tablets (B) and the model group (C), ten rabbits each group. The rabbit received intragastric administration of GuBi ZhiTong liquid, glucosamine hydrochloride tablets and physiological saline for eight weeks consecutively after establishing the rabbit model with knee osteoarthritis. Morphological changes were observed by the naked eyes, the expressions of IL-1 and TNF-α in the cartilage were detected by HE staining and RT-PCR after the samples were taken in eight weeks. Results: The observation under the naked eyes: there was no obvious change in the comparison of A and B between before and after medication, the damage of joint cartilage was severe in group C, even erosion. RT-PCR: when A and B were compared with C, the expressions of IL-1mRNA and TNF-αmRNA lowered obviously in A and B (P＜0.05). The expressions of IL-1 mRNA and TNF-α mRNA in the rabbit cartilage of A were lower than these of B (P＜0.05). Conclusion: GuBi ZhiTong liquid could inhibit inflammatory reaction by reducing the expressions of IL-1 and TNF-α, therefore, to relieve joint damage.
