目的:观察参茸益精片对急性高原缺氧脑损伤的保护作用,并探讨其分子机制。方法:将18只SD大鼠随机分为空白对照组,模型对照组和参茸益精片治疗组,每组各6只,建立急性高原缺氧脑损伤大鼠动物模型,空白对照组大鼠给予常规饲料喂养,自由饮水,饲养环境维持在温度25℃和湿度50%,明暗周期12小时/12小时,模型对照组大鼠在上述饲养环境中,给予常规饲料并自由饮水,而治疗组动物在给予上述处理的同时,口服灌胃参茸益精片0.5 mg/kg。8周后,观察大鼠脑组织中Caspase-3和Caspase-9活性的影响,并通过Western blot和RT-PCR等实验,观察Caspase-3和Caspase-9蛋白和mRNA表达的变化。结果:与空白对照组相比,急性高原缺氧造成大鼠脑损伤,引起Caspase-3和Caspase-9活性显著增加(P<0.05,n=6),分子生物学结果也显示,其表达水平显著增加(P<0.05,n=6);参茸益精片治疗组的动物,Caspase-3和Caspase-9显著降低(P<0.05,n=6)。结论:参茸益精片可显著增加大鼠脑组织中Caspase-3和Caspase-9的活性和表达水平,可能与其脑保护作用有关。

Objective: To observe the protective effects of ShenRong YiJing tablets against acute high altitude anoxia cerebral injury, and discuss its molecular mechanism. Methods: All 18 rats were randomized into blank control group, model control group and the treatment group, six rats each group.Animal models with acute high altitude anoxia cerebral injury were established, the rats in blank control group were fed with conventional feedstuff, drink-ing water freely, the rearing environment maintained at 25℃ and 50% humidity,light and dark period: 12 hours/12 hours, the rate in model control group were given conventional feedstuff, drinking water freely, the animals in the treatment group took ShenRong YiJing tablets based on the environment, the effects of the medicine on the activity of Caspase-3 and Caspase-9 in cerebral tissue of the rats were observed, and Caspase-3 and Caspase-9 protein and mRNA expressions changes were observed by western blot, RT-PCR and others. Results: Compared with normal control group, acute high altitude anoxia made cerebral injury of the rats, which could increase the activity of Caspase-3 and Caspase-9 significantly (P<0.05, n=6), molecular biological results indicated that the expressions rose notably (P<0.05, n=6); while the expressions of Caspase-3 and Caspase-9 decreased remarkably of the animals in the treatment group of ShenRong YiJing tablets (P<0.05, n=6). Conclusion: ShenRong YiJing tablets could notably increase the activity and the expressions of Caspase-3 and Caspase-9 in cerebral tissue of the rats, which might be related to cerebral protection.
