摘要:
目的：探讨大蒜素对链脲佐菌素所致糖尿病大鼠胰岛素抵抗及脂肪因子的影响。方法：通过高脂高糖饮食结合腹腔注射链脲佐菌素的方法建立糖尿病大鼠模型并给予不同剂量大蒜素［5、10、20 mg/(kg·d)］和盐酸二甲双胍［200 mg/(kg·d)］进行治疗，1次/d，疗程4周，另设模型组和正常对照组。给药前和给药后第7、14、21、28天测定大鼠空腹血糖(fasting blood glucose，FBG)水平和空腹胰岛素(fasting serum lisulin，FINS)水平；计算4周后胰岛素敏感指数(Insulin sensitivity index，ISI)及胰岛素抵抗指数(HOMA-IR)，测定血清血脂指标和血浆中脂肪因子水平。结果：与模型组比较大蒜素组大鼠FBG水平降低，FINS水平升高，HOMA-IR及ISI降低；总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)含量降低而高密度脂蛋白胆固醇(HDL-C)含量升高；瘦素(Leptin)、肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)降低，脂联素(Adiponectin)水平升高，且具有一定剂量依赖性，其中大蒜素20 mg/(kg·d)组效果最显著。结论：大蒜素对链脲佐菌素所致糖尿病大鼠具有剂量依赖性改善胰岛素抵抗和调节脂肪因子含量的作用，其作用优于盐酸二甲双胍。
Abstract:
Objective: To explore the effects of allitridi on insulin resistance and adipokines of the rats with streptozotocin(STZ)-induced diabetes mellitus(DM). Methods: DM rat models were prepared by high fat and high sugar diet and abdominal injection of STZ, they were given different concentrations of allitridi[5, 10 and 20mg/(kg·d)] and deltamine[200 mg/(kg·d)], once each day, the course was four weeks, the model group and normal control group were set up. The levels of fasting blood glucose (FBG) and fasting serum insulin(FINS) of the rats were detected before medicating, at the seventh, 14 th, 21st and 28 th day after medicating; insulin sensitivity index(ISI) and HOMA-IR were calculated in four weeks, the levels of th indexes of blood lipid and adipokines were detected. Results: Compared with the model group, the levels of FBG decreased while the levels of FINS rose in the allitridi group; HOMA-IR and ISI lowered; the contents of TC, TG and LDL-C lowered while the contents of HDL-C increased; the levels of leptin, TNF-α and IL-6 lowered while the level of adiponectin raised, which were in a dose-dependent manner, and the effects of allitridi in the concentrations of 20 mg/(kg·d)] were the most obvious. Conclusion: Allitridi could improve insulin resistance and regulate the contents of adipokines of the rats with STZ-induced DM in a dose-dependent manner, and its effects are superior to these of deltamine.
