目的通过分析姜黄素对库普夫细胞(kupffer cells,KCs)促炎因子和趋化因子分泌功能的抑制情况,阐述姜黄素对急性肝损伤的治疗作用。 方法通过原位灌注和梯度离心的方法提取小鼠肝脏中原代KCs细胞,予以不同浓度的姜黄素预处理后,加入脂多糖(lipopolysaccharide,LPS)后培养24 h。实时定量PCR法检测KCs中促炎细胞因子肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、白细胞介素1β(interleukin-1β,IL-1β)、白细胞介素6(interleukin-6,IL-6)mRNA的表达及趋化因子CXC家族趋化因子配体1(C-X-C Motif Chemokine Ligand 1,CXCL1)、趋化因子CXC家族趋化因子配体2(C-X-C Motif Chemokine Ligand 2,CXCL2)、单核细胞趋化蛋白1(monocyte chemotactic protein 1,MCP-1)、单核细胞趋化蛋白2(monocyte chemotactic protein 2,MCP-2)和MCP-1的mRNA表达;利用WB检测ERK1/2、P38和JNK细胞通路的变化。 结果予以LPS刺激后,KCs表达的促炎细胞因子TNF-α、IL-1β、IL-6、CXCL1、CXCL2和MCP-1表达明显升高,予以不同浓度的姜黄素预处理后,TNF-α、IL-1β、IL-6、CXCL1、CXCL2和MCP-1表达均明显降低,且呈剂量依赖性,不同组别之间差异有统计学意义(P<0.05)。WB结果显示姜黄素可以缓解LPS诱导的ERK1/2、P38和JNK蛋白磷酸化水平(P<0.05)。 结论姜黄素可能是通过抑制KCs活化来减少促炎因子和趋化因子分泌,进而缓解急性肝损伤。

ObjectiveBy discussing the inhibitory effects of curcumin on the secretion of proinflammatory factors and chemokines of KCs, to further elucidate therapeutic effects of curcumin for acute liver injury. MethodsPrimary KCs cells were isolated from mice liver by in situ perfusion and gradient centrifugation, pretreated with different concentrations of curcumin and cultured for 24 hours after adding LPS. To detect the expressions of proinflammatory factors including TNF-α, IL-1β and IL-6 mRNA, and chemokines including CXCL1, CXCL2, MCP-1, MCP-2 and MCP-1 mRNA of KCs using real time quantitative PCR; WB was used to measure the changes of ERK1/2, P38 and JNK pathway. ResultsAfter LPS stimulation, the expressions of TNF-α, IL-1β, IL-6, CXCL1, CXCL2 and MCP-1 of KCs raised notably, after preconditioning with different concentrations of curcumin, the expressions of TNF-α, IL-1β, IL-6, CXCL1, CXCL2 and MCP-1 lowered remarkably, in the dose-dependent manner, and the differences were statistically significant between different groups (P<0.05). WB results displayed that curcumin could alleviate LPS-induced phosphorylation of ERK1/2, P38 and JNK proteins (P<0.05). ConclusionCurcumin could reduce the secretion of proinflammatory factors and chemokines which might be through inhibiting the activation of KCs, therefore to relieve acute liver injury.
