摘要:
目的探讨温肾化痰方对氧化低密度脂蛋白(oxidized low density lipoprotein,ox-LDL)诱导的动脉粥样硬化模型大鼠主动脉血管平滑肌细胞凋亡及细胞内胆固醇酯的影响。 方法将大鼠主动脉血管平滑肌细胞分为正常组(10% Gibco)、对照组(ox-LDL 70 mg/L+10% Gibco)及低剂量(ox-LDL 70 mg/L+温肾化痰方5.256 g/kg)、中剂量(ox-LDL 70 mg/L+温肾化痰方10.053 g/kg)、高剂量中药组(ox-LDL 70 mg/L+温肾化痰方20.106 g/kg)。MTT法检测大鼠主动脉血管平滑肌细胞存活率。流式细胞术(FCM)检测大鼠主动脉血管平滑肌细胞凋亡水平;荧光分析法测定各组细胞内胆固醇及胆固醇酯含量。 结果与正常组比较,对照组主动脉平滑肌细胞存活率下降,凋亡率升高,总胆固醇及胆固醇酯水平升高(均P<0.01);与对照组比较,各中药组主动脉平滑肌细胞存活率增加,凋亡率降低,总胆固醇及胆固醇酯水平降低(P<0.01);与低剂量中药组比较,中、高剂量中药组主动脉平滑肌细胞存活率增加,凋亡率降低(P<0.01),总胆固醇及胆固醇酯水平降低(P<0.01);与中剂量中药组比较,高剂量中药组主动脉平滑肌细胞存活率增加,凋亡率降低(P<0.01),总胆固醇及胆固醇酯水平降低(P<0.01),呈现浓度依赖性。 结论温肾化痰方可抑制ox-LDL诱导的动脉粥样硬化模型大鼠主动脉血管平滑肌细胞凋亡,提高存活率;降低总胆固醇及胆固醇酯水平,保护血管平滑肌细胞,延缓动脉粥样硬化进程。
Abstract:
ObjectiveTo explore the effects of kidney-warming phlegm-resolving prescription (KWPRP) on the apoptosis of aortic vascular smooth muscle cells and intracellular cholesterol esters on ox-LDL-induced atherosclerosis rat models. MethodsRat aortic vascular smooth muscle cells were divided into the normal group (10% Gibco), the control group (ox-LDL 70 mg/L+10% Gibco), low dose group (ox-LDL 70 mg/L+KWPRP 5.256 g/kg), moderate dose group (ox-LDL 70 mg/L+KWPRP 10.053 g/kg) and high dose group of herbs (ox-LDL 70 mg/L+KWPRP20.106 g/kg), the survival rate of rat aortic vascular smooth muscle cells was detected by MTT assay. The apoptosis of rat aortic vascular smooth muscle cells was measured by flow cytometry (FCM); the contents of intracellular cholesterol and cholesterol ester in different groups were surveyed by fluorescence analysis. ResultsCompared with the normal group, the survival rate of aortic vascular smooth muscle cells decreased, the apoptosis rate lifted, the levels of total cholesterol and cholesterol ester ascended in the control group (all P<0.01); compared with the control group, the survival rate of aortic vascular smooth muscle cells increased, the apoptosis rate lowered, and the levels of total cholesterol and cholesterol ester decended in all the herb groups (P<0.01); compared with low dose group of herb, the survival rate of aortic vascular smooth muscle cells increased, and the apoptosis rate lowered in moderate and high dose groups of herbs (P<0.01), the levels of total cholesterol and cholesterol ester decreased (P<0.01); compared with moderate dose group of herb, the survival rate of aortic vascular smooth muscle cells rose, and the apoptosis rate lowered in high dose group of herbs (P<0.01), the levels of total cholesterol and cholesterol ester reduced (P<0.01), presenting in a dose-dependent manner. ConclusionKidney-warming phlegm-resolving prescription could inhibit the apoptosis of aortic vascular smooth muscle cells in ox-LDL - induced atherosclerosis rat models, lift survival rate; reduce the levels of total cholesterol and cholesterol ester, protect vascular smooth muscle cells and delay the progression of atherosclerosis.
