摘要:
目的基于网络药理学方法探讨蒲黄治疗糖尿病视网膜病变(diabetic retinopathy,DR)的作用机制。 方法运用中药系统药理分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)提取蒲黄的活性成分及对应靶点,借助UniProt数据库对靶点进行规范化,通过GeneCards数据库和OMIM数据库获取DR的相关靶点,并利用R语言筛选药物与疾病的交集靶点。通过Cytoscape软件构建蒲黄“活性成分-疾病靶点”可视化调控网络,利用STRING数据库构建蛋白互相作用网络,并借助Bioconductor平台进行基因本体论(gene ontology,GO)富集分析及京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)富集通路分析。 结果共获得蒲黄6个活性成分,药物与疾病127个交集靶点,核心靶点包含蛋白激酶B、血管内皮生长因子A、肿瘤蛋白53等;GO功能富集分析涉及转录因子活性、酶活性、受体活性及生物化学过程调控等过程,KEGG通路富集分析涉及动脉粥样硬化、AGEs-RAGE等通路。 结论蒲黄中的槲皮素等活性成分作用于Akt1等疾病靶点,参与酶结合、应激反应等生物学功能调控AGEs-RAGE等信号通路,从而治疗DR。
Abstract:
ObjectiveTo investigate the mechanism of treating diabetic retinopathy (DR) with Puhuang (typhae pollen) based on network pharmacology. MethodsTCMSP was used to extract the active ingredients and the corresponding targets of Puhuang, the standardization of the targets was conducted via Uniprot database, DR-related targets were obtained from GeneCards database and OMIM database, the intersecting targets between medicine and disease were screened using R language; visualization of "active ingredients-disease targets" control network of Puhuang was constructed via Cytoscape software, protein interaction network was built using STRING database, GO enrichment analysis and KEGG enrichment pathway was performed via Bioconductor platform. ResultsThe study has yielded six active ingredients, and 127 intersecting targets between the medicine and disease, core targets contained protein kinase B, vascular endothelial growth factor A, tumour protein 53 and others; GO functional enrichment analysis involve processes such as the activity of transcription factor, enzyme and receptor, and regulation of biochemical processes, and KEGG pathway enrichment analyses involve pathways such as atherosclerosis, AGEs-RAGE, etc. ConclusionActive ingredients such as quercetin in Puhuang could act on the disease targets such as Akt1 and participate in biological functions including enzyme binding, and stress response, to regulate AGEs-RAGE signaling pathway, thereby treating DR.
