摘要:
目的探讨左归丸对60Coγ射线损伤SD大鼠外周血细胞、性激素水平和白细胞介素1β(Interleukin 1β,IL-1β)水平及卵巢卵泡数量、脾脏指数的影响。 方法选择8周龄雌性SPF级SD大鼠50只,其中10只为正常组,不进行照射,常规饲养;其余40只以60Coγ射线一次性照射(6.0Gy,LD40)24 h后,分为辐射模型组,补佳乐组,左归丸高、低剂量组。补佳乐组予补佳乐0.09 mg;左归丸高剂量组予左归丸4.725 g生药和左归丸低剂量组予左归丸0.945 g生药;辐射组予生理盐水2 mL灌胃,每日1次,共干预21天。检测不同时间段外周血细胞变化情况;酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)检测外周血血清中卵泡刺激素(follicle stimulating hormone,FSH)、雌激素(estradiol,E2)、黄体生成素(luteinizing hormone,LH)和白细胞介素1β(Interleukin 1β,IL-1β)表达水平,并测定卵巢卵泡数量及脾脏指数。 结果辐射后大鼠外周血细胞数量减少,尤以白细胞减少最显著(P<0.05),血红蛋白含量增加(P<0.05),E2水平降低,卵巢内成熟卵泡数量减少,脾脏指数降低,IL-1β表达水平升高;左归丸干预后大鼠血清E2水平成熟卵泡数量及脾脏指数升高,血清IL-1β、FSH、LH水平均降低,尤以左归丸高剂量组作用更明显(P<0.05)。 结论左归丸能够促进辐射损伤大鼠卵巢功能修复,可能与促进骨髓造血,提高免疫功能有关,体现了“肾主血脉”理论。
Abstract:
ObjectiveTo discuss the influence of Zuogui pills on the levels of peripheral blood cells and sex hormone and IL-1β, the number of ovarian follicles as well as spleen indexes in SD rats suffering 60Coγ-rays injury. MethodsA total of 50 female SPF grade SD rats at eight weeks of age were selected, ten of them were in the normal group, not irradiated and routinely reared; the rest 40 were divided into radiation model group, estradiol valerate tablets group, high and low dosage groups of Zuogui pills after irradiated with 60Coγ-rays (6.0Gy, LD40) for 24 h. Estradiol valerate tablets were administered to estradiol valerate tablets 0.09 mg; high and low dosage groups of Zuogui pills were given Zuogui pills at the concentrations of 4.725 g crude drug and 0.945 g crude drug; radiation model group was drenched with physiological saline 2 mL, once each day, and intervened for 21 days. To detect the changes of peripheral blood cells at different time intervals; ELISA was used to detect the expressions of FSH, E2, LH and IL-1β in peripheral blood, to measure the number of ovarian follicles and spleen indexes. ResultsAfter being irridated, the number of peripheral blood cells was decreased, and the reduction of white blood cells was most pronounced (P<0.05), the contents of hemoglobin were increased (P<0.05), the level of E2 was lowered, the number of mature follicles was reduced, spleen index was lowered, and the expressions of IL-1β were lifted; after the intervention of Zuogui pills, the levels of E2, the number of mature follicles and spleen indexes were increased, the levels of IL-1β, FSH and LH were lowered, especially the effects were more pronounced in the high-dose group of Zuogui pills (P<0.05). ConclusionZuogui pills could promote the restoration of ovarian function in rats with radiation damage possibly through promoting bone marrow hematopoiesis and immune function, which reflects the theory of "kidney governing the blood vessels".
