摘要:
目的探讨杜仲腰痛丸对大鼠椎间盘软骨终板细胞的作用机制。 方法使用区组随机法将50只实验大鼠分为对照组、模型组、扶他林组、杜仲腰痛丸高、低剂量组5组,每组各10只。运用病因模拟造模方法成功复制腰椎间盘突出症大鼠模型,然后根据不同组别给予相应治疗。采集大鼠L6~7椎间盘软骨终板细胞组织标本,反转录实时荧光定量聚合酶链式反应PCR(reverse transcription-polymerase chain reaction,RT-PCR)法检测椎间盘软骨终板细胞组织标本中与椎间盘退变相关因子,转化生长因子β1(transforming growth factor-β1,TGF-β1)、Smad2(drosophila mothers against decapentaplegic protein 2)、Smad4、同源盒B4(homeobox B4,HOXB4)、Ⅺ型胶原蛋白α1链mRNA(Ⅺ type collagen,COL11A1mRNA)的表达量,蛋白质免疫印迹法(Western blot,WB)检测与椎间盘退变相关的TGF-β1、Smad2、Smad4、HOXB4、COL11A1蛋白表达,HE染色观察椎间盘软骨组织的病理变化情况。 结果模型组大鼠椎间盘软骨中TGF-β1、Smad2、Smad4、HOXB4、COL11A1 mRNA相对表达量均较空白对照组降低(P<0.01);经扶他林组治疗后,大鼠椎间盘TGF-β1、COL11A1、Smad2的mRNA相对表达量较模型组升高(P<0.05);经杜仲腰痛丸合剂治疗后,大鼠椎间盘TGF-β1、Smad2、Smad4、HOXB4、COL11A1的mRNA相对表达量高于模型对照组(P<0.05);高剂量组与低剂量组比较,高剂量组TGF-β1、Smad2、HOXB4、COL11A1的mRNA相对表达量高于低剂量组(P<0.05)。从蛋白表达来看,与空白对照组比较,模型组大鼠椎间盘软骨TGF-β1、Smad2、HOXB4、COL11A1蛋白相对表达量均升高(P<0.05);经扶他林组治疗后TGF-β1、Smad4蛋白相对表达量高于模型组(P<0.05);高剂量杜仲腰痛丸组治疗后TGF-β1、Smad2、Smad4、HOXB4、COL11A1蛋白相对表达量较模型组升高(P<0.05);低剂量杜仲腰痛丸组与模型组比较,低剂量杜仲腰痛丸组的TGF-β1、Smad2、Smad4、COL11A1蛋白表达量相对升高(P<0.05)。 结论杜仲腰痛丸合剂可抑制软骨终板细胞的凋亡,对椎间盘软骨终板细胞具有保护作用,其相关分子生物学机制可能与调节TGF-β1/Smad2/Smad4-HOXB4/COL11A1信号通路相关。
Abstract:
ObjectiveTo discuss the effects of Duzhong Yaotong pills on rat intervertebral disc cartilage endplate cells. MethodsFifty rats were randomized into five groups including the control group, the model group, diclofenac sodium (voltaren) group, high and low dosages groups of Duzhong Yaotong pills (n=10 in each group). Rat models suffering lumbar disc herniation (LDH) were duplicated successfully using etiological simulation modelling methods, and they accepted the corresponding therapy in different groups. After collecting the samples of rat intervertebral disc cartilage endplate cells from L6-7, RT-PCR was applied to detect the expressions of the factors related to intervertebral disc degeneration, TGF-β1 , Smad2 and Smad4, HOXB4 and COL11A1mRNA in the samples, WB was applied to measure the expressions of the protein related to intervertebral disc degeneration such as TGF-β1 , Smad2 and Smad4, HOXB4 and COL11A1 protein, and HE staining to observe the pathological changes of intervertebral disk cartilage. ResultsThe relative expressions of TGF-β1 , Smad2 and Smad4, HOXB4 and COL11A1mRNA were lowered in the model group than blank control group (P<0.01); after treatment with diclofenac sodium, the relative expressions of TGF-β1 , COL11A1 and Smad2 mRNA were elevated than the model group (P<0.05); the relative expressions of TGF-β1 , Smad2 and Smad4, HOXB4 and COL11A1 mRNA in the intervertebral disc were higher after the treatment with the mixture of Duzhong Yaotong pills than these of model control group (P<0.05); the relative expressions of TGF-β1 , Smad2, HOXB4 and COL11A1 mRNA in the high dosage group were higher than these in the low dosage group (P<0.05). In terms of the expressions of the protein, compared with blank control group, the relative expressions of TGF-β1 ,Smad2, HOXB4 and COL11A1 protein were higher in the model group (P<0.05); after diclofenac sodium treatment, the relative expressions of TGF-β1 and Smad4 were higher than these of the model group (P<0.05); the relative expressions of TGF-β1, Smad2 and Smad4, HOXB4 and COL11A1 protein in the high dose group of the medicine were increased than these of the model group (P<0.05); when low dosage group of the medicine was compared with the model group, the relative expressions of TGF-β1 ,Smad2 and Smad4, and COL11A1 protein were higher (P<0.05). ConclusionThe mixture of Duzhong Yaotong pills could inhibit the apoptosis of cartilage endplate cells and protect intervertebral disc cartilage endplate cells, and the related molecular mechanism might be related to the regulation of TGF-β1 /Smad2/Smad4-HOXB4 /COL11A1 signaling pathway.
