摘要:
目的探讨沉默LncRNA-H19联合小剂量冬凌草甲素对急性早幼粒细胞白血病(acute promyelocytic leukemia,APL)NB4细胞增殖和凋亡的影响。 方法体外常规培养NB4细胞,通过转染LncRNA-H19 siRNA沉默LncRNA-H19表达,将细胞分为对照组、LncRNA-H19 siRNA组、冬凌草甲素组和联合干预组。采用RT-PCR法检测细胞中LncRNA-H19表达,CCK-8法检测细胞增殖抑制率、流式细胞仪检测细胞凋亡率、Western Blot法检测细胞中目的蛋白表达水平。 结果LncRNA-H19 siRNA组和联合干预组细胞中LncRNA-H19表达量显著低于对照组(P<0.05),细胞增殖抑制率和凋亡率显著高于对照组(P<0.05),细胞中β-catenin、Cyclin D1、C-myc、PI3K和AKT蛋白表达量明显低于对照组(P<0.05);冬凌草甲素组细胞增殖抑制率和凋亡率明显高于对照组(P<0.05),细胞中PI3K蛋白表达量明显低于对照组(P<0.05);联合干预组干预效果明显优于LncRNA-H19 siRNA组或冬凌草甲素组(P<0.05)。 结论沉默LncRNA-H19联合小剂量冬凌草甲素能显著抑制APL进展,可能与其抑制Wnt/β-catenin和PI3K/AKT信号通路及其下游因子有关。
Abstract:
ObjectiveTo discuss the effects of silent LncRNA-H19 combined with small dose oridonin on the proliferation and apoptosis of APL NB4 cells. MethodsNB4 cells were routinely cultured in vitro, the cells were divided into the control group, LncRNA-H19 siRNA group, oridonin group and joint intervention group after silencing the expressions of LncRNA-H19 by transfection of LncRNA-H19 siRNA. RT-PCR method was used to detect the expressions of LncRNA-H19, CCK-8 method to measure cell proliferation inhibition rate, flow cytometry to detect cellular apoptosis rate, Western Blot method to measure the expressions of target protein in cells. ResultsThe expressions of LncRNA-H19 in LncRNA-H19 siRNA group and joint intervention group were lower than these of the control group (P<0.05), cell proliferation inhibition rate and the apoptosis rate were higher than these of the control group (P<0.05), the expressions of β-catenin, Cyclin D1, C-myc, P13K and AKT protein were lower than these of the control group (P<0.05); cell proliferation inhibition rate and the apoptosis rate of the oridonin group were higher than these of the control group (P<0.05), the expressions of PI3K protein in the cells were lower than these of the control group (P<0.05); the intervention effects of joint intervention group were notably better than these of LncRNA-H19 siRNA group or oridonin group (P<0.05). ConclusionSilent LncRNA-H19 combined with small dose oridonin could notably inhibit the progression of APL, which might be related to that it could inhibit Wnt/β-catenin and PI3K/AKT signaling pathway and its downstream factors.
