摘要:
目的探讨紫草素对宫颈癌细胞株Caski细胞侵袭迁移及Wnt/β-catenin信号通路的影响。 方法采用MTT法检测细胞增殖抑制情况;采用Transwell小室侵袭迁移实验检测Caski细胞侵袭迁移情况;采用qPCR方法检测Caski细胞GSK-3β、Wnt2B表达情况;采用Western bloting方法检测Caski细胞β-Catenin、p-β-Catenin蛋白含量。 结果紫草素可抑制Caski细胞的增殖,具有时间-浓度依赖性,并可抑制细胞的侵袭迁移,增加Caski细胞GSK-3β mRNA和p-β-Catenin蛋白的表达量,减少Caski细胞Wnt mRNA和β-Catenin蛋白的表达量。 结论紫草素可能通过抑制Caski细胞Wnt/β-Catenin信号通路抑制Caski细胞的侵袭迁移。
Abstract:
ObjectiveTo explore the effects of shikonin on the invasion and migration of cervical cancer cell line Caski and Wnt/β-catenin signaling pathway. MethodsMTT method was used to detect the inhibition of cell proliferation; Transwell chamber invasion and migration experiment was adopted to measure the invasion and migration of Caski cells; qPCR method to measure the expressions of GSK-3β and Wnt2B in Caski cells; Western bloting method to measure the contents of β-Catenin and p-β-Catenin protein in Caski cells. ResultsShikonin could inhibit the proliferation of Caski cells, in a time-concentration dependent manner, it could inhibit cellular invasion and migration, in crease the expressions of GSK-3β mRNA and p-β-Catenin protein, and lower the contents of Wnt mRNA and β-Catenin protein in Caski cells. ConclusionShikonin could restrain the invasion and migration of Caski cell possibly through inhibiting Wnt/β-catenin signaling pathway.
