摘要:
目的探讨葛根素调节钙调素依赖性蛋白激酶Ⅱ(CaMKⅡ)对布比卡因中毒大鼠心肌损伤的影响。 方法将雄性Wistar大鼠40只采用随机数字表法分为空白对照组(C组)、布比卡因中毒模型组(B组)、葛根素预防组(P组)、葛根素治疗组(T组),每组10只。C组经股静脉输注0.9%生理盐水3 mL/kg;B组经股静脉输注0.5%布比卡因10 mg/kg,出现心律失常后输注0.9%生理盐水3 mL/kg;P组经股静脉输注葛根素3 mL/kg 5 min后输注0.5%布比卡因10 mg/kg;T组经股静脉输注0.5%布比卡因10 mg/kg,出现心律失常后立即输注葛根素3 mL/kg。C组在开始输注生理盐水的第8 min及B、T、P组在开始输注利多卡因的第8 min,处死大鼠。取左室心肌组织,采用流式细胞仪测定心肌细胞Ca2+浓度,Western blot法测定心肌细胞CaMKⅡ磷酸化水平,TUNEL染色观察细胞凋亡情况。 结果与C组比较,B组心肌细胞CaMKII磷酸化水平、Ca2+浓度、细胞凋亡率均升高(P<0.05)。与B组比较,P组心肌细胞CaMKII磷酸化水平、Ca2+浓度、细胞凋亡率均降低(P<0.05),T组心肌细胞CaMKⅡ磷酸化水平、Ca2+浓度、细胞凋亡率均降低,但差异无统计学意义(P>0.05)。 结论葛根素可能通过调节CaMKⅡ来减轻布比卡因中毒大鼠的心肌损伤。
Abstract:
ObjectiveTo investigate the effect of puerarin on myocardial damage in bupivacaine intoxicated rats via regulating CaMKⅡ. MethodsForty male Wistar rats were allocated to blank control group (C group), bupivacaine intoxicated model group (B group), puerarin prevention group (P group) and puerarin treatment group (T group) using random number table method, with ten rats in each group. C group was infused with 0.9% physiological saline, 3 mL/kg, through femoral vein; B group was infused with 0.5% bupivacaine, 10 mg/kg, through femoral vein, and 0.9% physiological saline, 3 mL/kg, when cardiac arrhythmia occurred; P group with puerarin, 3 mL/kg, through femoral vein, and 0.5% bupivacaine, 10 mg/kg, in five minutes; T group with 0.5% bupivacaine, 10mg/kg, through femoral vein, and puerarin, 3 mL/kg, instantly when cardiac arrhythmia occurred. The rats were sacrificed at the eighth minute when C group began to infuse physiological saline, B group, T group and P group began to infuse lidocaine. To detect the concentrations of myocardial Ca2+ using flow cytometry after taking left ventricular myocardial tissue, to detect phosphorylation level of myocardial CaMKⅡ using Western blot method, to observe cellular apoptosis using TUNEL staining. ResultsCompared with C group, phosphorylation level of myocardial CaMKⅡ, the concentrations of Ca2+ and cellular apoptosis rate of B group rose (P<0.05). Compared with B group, phosphorylation level of myocardial CaMKⅡ, the concentrations of Ca2+ and cellular apoptosis rate of P group decreased (P<0.05), phosphorylation level of myocardial CaMKⅡ, the concentrations of Ca2+ and cellular apoptosis rate of T group lowered, while the difference had no statistical meaning (P>0.05). ConclusionPuerarin could relieve myocardial damage in bupivacaine intoxicated rats possibly through regulating the levels of CaMKⅡ.
