摘要:
目的探讨健胃清肠合剂对不完全性肠梗阻大鼠肠黏膜免疫屏障功能的影响。 方法按照随机数字表法将112只SPF级Wistar雄性大鼠分为正常组,假手术组,模型组,阳性对照组,健胃清肠高、中、低剂量组7组,每组16只。除假手术组及正常组外,其余各组以结扎法复制不完全性肠梗阻大鼠模型。健胃清肠各剂量组大鼠分别以高、中、低浓度的健胃清肠合剂混合液灌胃;阳性对照组予石蜡油-蒸馏水混合液灌胃;其余3组予蒸馏水灌胃,共干预3天。采用酶联免疫法(enzyme-linked immuno sorbent assay,ELISA)检测血清中白细胞介素6(interleukin-6,IL-6)、白细胞介素10(interleukin-10,IL-10)及α-干扰素(interferon-α,IFN-α)的表达水平。 结果与模型组比较,健胃清肠合剂各剂量组血清中IL-6、IL-10及IFN-α的水平均降低,尤以高、中剂量组降低更明显(P<0.05);且健胃清肠合剂高、中剂量组大鼠血清中IL-6、IL-10及IFN-α的水平相比与阳性对照组相比,差异有统计学意义(P<0.05)。 结论健胃清肠合剂能够有效降低不完全性肠梗阻大鼠血清中炎性因子IL-6、IL-10及IFN-α的表达水平,对大鼠肠黏膜免疫屏障起保护作用。
Abstract:
ObjectiveTo explore the effects of Jianwei Qingchang mixture (JWQCM) on intestinal mucosal immune barrier function in rats with incomplete intestinal obstruction. MethodsAll 112 SPF Wistar male rats were divided into the normal group, sham operation group, the model group, positive control group, high, moderate and low dose groups of JWQCM according to random number table method, 16 rats in each group. Except sham operation group and the normal group, the rats in other groups were duplicated into rat models with incomplete intestinal obstruction by ligation therapy. Different dosage groups of JWQCM were drenched with high, moderate and low concentrations of JWQCM respectively; and positive control group with the mixture of paraffin oil and distilled water; and the other groups with distilled water, intervened for three days altogether. ELISA method was adopted to detect the expressions of IL-6, IL-10 and IFN-a in the serum. ResultsCompared with the model group, the levels of IL-6, IL-10 and IFN-a in the serum of different doses groups of JWQCM lowered, especially the decrease of high and moderate doses groups was noticeable (P<0.05); when the levels of IL-6, IL-10 and IFN-a in high and moderate doses groups of JWQCM were compared with these in positive control group, the difference showed statistical meaning (P<0.05). ConclusionJWQCM could effectively reduce the expressions of IL-6, IL-10 and IFN-a in the serum of the rats with incomplete intestinal obstruction, and it could protect intestinal mucosal immune barrier in rats.
