摘要:
目的基于网络药理学原理探讨消疕汤核心药对金银花-连翘干预银屑病的分子通路及作用机制。 方法应用中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmaco-logy database and analysis platform,TCMSP)检索金银花、连翘化学成分、作用靶点,并通过GeneCards数据库获取银屑病差异性靶点基因,预测中药与银屑病相关的作用靶点,通过STRING平台构建蛋白质-蛋白质相互作用信息网络(PPI),运用DAVID数据库对筛选出的靶点基因进行基因富集分析,获得基因本体论功能富集分析(gene ontology,GO)和KEGG富集分析(kyoto encyclopedia of genes and genomes,KEGG)信号通路注释结果,构建“中药成分-关键靶标-主要通路”的可视化网络图,探究金银花-连翘药对干预银屑病的分子作用机制。 结果金银花-连翘药对干预银屑病,共预测得到28个活性成分,涉及核心靶标基因127个,生物功能138个和信号通路155个。 结论金银花-连翘药对主要活性成分通过多靶点、多通路干预银屑病,主要涉及核受体的活性、转录因子活性、细胞因子受体结合、细胞因子生物功能等和白细胞介素17(interleukin-17,IL-17)信号通路、肿瘤坏死因子(tumor necrosis factor,TNF)信号通路、磷脂酰肌醇3-激酶(phosphatidylinositol 3-kinase,PI3K)-蛋白激酶B(protein kinase B,Akt)信号通路等。
Abstract:
ObjectiveTo discuss molecular pathway and mechanism of Xiaobi Tang core couplet medicine Jinyinhua (Lonicerae japonicae flos)-Lianqiao (Forsythiae fructus) in the intervention of psoriasis based on network pharmacology. MethodsChemical ingredients and targets of Jinyinhua and Lianqiao were retrieved from TCMSP, and differential target genes of psoriasis were obtained from GeneCards database, to predict psoriasis-related targets in the herbs, to construct PPI via STRING platform, gene enrichment of the screened targets were analyzed by using DAVID database, to obtain the annotation notes of GO biological function and KEGG signaling pathway, to construct visual network diagram of “herbal ingredients-key targets-main pathway” and explore the molecular mechanism of materia medica in the intervention of psoriasis. ResultsIn the intervention of psoriasis by couplet medicine Jinyinhua-Lianqiao, 28 active ingredients were gained by the prediction, involving 127 core target genes, 138 biological function and 155 signaling pathway. ConclusionJinyinhua-Lianqiao could intervene psoriasis by many targets and pathways, mainly involving the activity of nuclear receptor, the activity of transcription factors, cytokine receptor binding, cytokine biological function and others, as well as IL-17 signaling pathway, TNF signaling pathway and P13K-Akt signaling pathway.
