摘要:
目的探讨白杨素对心肌缺血再灌注损伤(myocardial ischemia reperfusion injury,MIRI)大鼠心肌组织环氧合酶2(cyclooxygenase-2,COX-2)和一氧化氮合酶(nitric oxide synthase,NOS)表达的影响。 方法将60只清洁级雄性SD大鼠按照随机数字表法分为假手术组、模型组、雷米普利组及白杨素低、中、高剂量组,每组10只,相应药物预防给药1周后构建MIRI模型,检测心肌病理学、血清心肌酶肌酸激酶(creatine kinase,CK)、肌酸激酶同工酶(creatine kinase isoenzyme,CK-MB)、乳酸脱氢酶(lactate dehydrogenase,LDH)和血浆肌钙蛋白I(troponin I,cTnI);检测心肌组织与血清炎性因子肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、白细胞介素1β(interleukin-1β,IL-1β)和白细胞介素10(interleukin-10,IL-10);采用蛋白印迹法(Western Blotting)和逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)方法检测心肌组织COX-2和NOS蛋白及其mRNA表达。 结果假手术组心肌纤维结构完整,排列整齐;模型组有明显纤维断裂,各治疗组相较于模型组心肌纤维结构明显好转,白杨素中、高剂量组效果好于雷米普利组。与假手术组相比,模型组心肌酶CK、CK-MB、LDH和cTnI以及心肌组织和血清中炎性因子TNF-α和IL-1β均升高(P<0.05),IL-10降低(P<0.05),各治疗组相较于模型组上述指标除IL-10升高外(P<0.05),其余指标均降低(P<0.05),白杨素中、高剂量组与雷米普利组降低心肌酶指标效果相当(P>0.05);白杨素低剂量组差于雷米普利组,且除cTnI均有统计学意义(P<0.05);对炎性因子影响与雷米普利组比较,除低剂量组对心肌组织TNF-α影响差于雷米普利组(P<0.05)外,其余各组差异均无统计学意义(P>0.05)。与假手术组相比,模型组心肌组织COX-2和NOS升高(P<0.05),相较于模型组各治疗组COX-2和NOS均降低(P<0.05),白杨素高剂量组降低COX-2表达效果优于雷米普利组(P<0.05)。 结论白杨素能够减轻MIRI大鼠心肌损伤,这与其可抑制心肌组织COX-2和NOS表达,进而降低炎症反应相关。
Abstract:
ObjectiveTo discuss the influence of chrysin on the expressions of COX-2 and NOS in myocardial tissue of rats with MIRI. MethodsSixty male SD rats were divided into sham operation group, the model group, ramipril group, low, moderate and high dosages groups of chrysin, with ten rats in each group, MIRI models were established one week after corresponding drug prophylaxis administration, to detect myocardial pathology, CK, CK-MB, LDH and cTn1; measure the levels of inflammatory factors TNF-α, IL-1β and IL-10 in serum and myocardium; Western Blotting and RT-PCR method were applied to test the expressions of COX-2 and NOS proteins and mRNA expressions in myocardial tissues. ResultsMyocardial fibres are structurally intact and neatly arranged in sham operation group; there was obvious fibre fracture in the model group, fibre structure was noticeably recovered in different therapeutic groups compared with the model group. The effects of moderate and high dose groups of chrysin were better than these of ramipril group. Compared with sham operation group, CK,CK-MB, LDH and cTn1 in myocardial enzyme and the levels of TNF-α and IL-1β were heightened in serum and myocardium of the model group (P<0.05), while the level of IL-10 was lowered (P<0.05), besides that the level of IL-10 was raised, when different therapeutic groups were compared with the model group, the other indexes were lowered (P<0.05). The effects were equivalent in lowering myocardial enzyme between moderate and high dose groups of chrysin and ramipril group (P>0.05); low dose group of chrysin was poorer than ramipril group, and there was statistical meaning except cTnI (P<0.05); when compared with ramipril group with the effects on inflammatory factors, and the effects on TNF-α in myocardial tissue in low dose group of chrysin were poorer than these of ramipril group (P<0.05), the difference had no statistical meaning in the remaining groups (P>0.05). The expressions of COX-2 and NOS were decreased in different therapeutic groups than these in the model group (P<0.05), the expressions of COX-2 of high dose group of chrysin were higher than these of ramipril group, and the difference had statistical meaning (P<0.05). ConclusionChrysin could relieve myocardial injury in MIRI rats, which is related to inhibiting the expressions of COX-2 and NOS in myocardial tissue, and consequently reduce inflammatory reaction.
