摘要:
目的基于网络药理学方法研究桂枝茯苓胶囊治疗盆腔炎的分子作用机制。 方法通过中药系统药理学数据库和分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)筛选桂枝茯苓胶囊的中药活性成分及作用靶点,通过人类基因综合数据库GeneCards寻找盆腔炎的作用靶点,两者取交集。通过Cytoscape 3.7.2软件构建药物-化学成分-靶点-疾病网络。通过蛋白互作String平台构建蛋白质互作网络(protein-protein interaction networks,PPI)并筛选核心基因。通过基因本体数据库Bioconductor对靶点基因进行基因本体论富集分析(gene ontology,GO)和KEGG(kyoto encyclopedia of genes and genomes,KEGG)富集分析。 结果共筛选出桂枝茯苓胶囊活性成分69个,涉及到177个作用靶点,从Genecards中获得4253个治疗盆腔炎的作用靶点,两者共有靶基因136个,关键靶点30个。GO富集分析主要涉及氧化应激反应、炎症反应、细胞凋亡等。KEGG通路富集主要包括晚期糖基化终末产物-晚期糖基化终末产物受体AGE-RAGE糖尿病并发症、前列腺癌、动脉粥样硬化、疱疹病毒感染、乙型肝炎、丙型肝炎、肿瘤坏死因子(tumor necrosis factor,TNF)、白细胞介素17(interleukin-17,IL-17)等信号通路。 结论桂枝茯苓胶囊通过多成分、多靶点、多通路发挥治疗盆腔炎的作用。
Abstract:
ObjectiveTo study the molecular mechanism of Guizhi Fuling capsules in the treatment of pelvic inflammatory disease (PID) on the foundation of network pharmacology. MethodsThe active ingredients and the targets of Guizhi Fuling capsules were screened from TCMSP, the targets of PID were investigated by GeneCards, and the intersection of both was taken out. Drug-chemical ingredients-targets-disease network were constructed via Cytoscape 3.7.2 software. PPI network was built and the core genes were screened out by String platform. GO and KEGG analysis of target genes were conducted by Bioconductor database. ResultsWe have screened out 69 active ingredients from Guizhi Fuling capsules, involving 177 targets, 4253 targets of treating PID from Genecards, 136 shared target genes between both and 30 key targets. GO enrichment analysis mainly contained oxidative stress response, inflammatory reaction, cellular apoptosis and others. KEGG pathway enrichment principally included AGE-RAGE diabetic complications signaling pathway, prostatic cancer pathway, atherosclerosis pathway, herpesvirus infection pathway, hepatits B pathway, hepatits C pathway, TNF signaling pathway and IL-17 signaling pathway. ConclusionGuizhi Fuling capsules could treat PID by multiple ingredients, multiple targets and multiple pathways.
